| Abstract: | Objective To study side effects of the anticancer drug hydroxycamptothecin on PC12 cell and discuss the side effect of HCPT on nervous system.Methods The growth inhibition was studied by CCK-8 assay.The morphologic changes of the apoptotic cells were observed by acridine orange/ethidium bromide dying.DNA of the apoptotic cells was analyzed with agarose gel electrophoresis（AGE）.The apoptosis rates and the cell cycle distribution were studied by flow cytometry after treatment with HCPT.Results CCK-8 assay showed that various concentration of HCPT could effectively inhibit the growth of cell line PC12.DNA ladder was observed in ultraviolet light.The typical apoptotic cells under the fluorescence microscope showed budding phenomena and apoptotic bodies.According to the results of flow cytometry,apoptosis rates increased with the treatment time prolonged under the same HCPT concentration.Conclusion HCPT has side effects and toxic action on PC12 cell.The effect is maybe related with its role of inducing cell apoptosis which might be correlated with distribution of the cell cycle. |
