柴胡疏肝散及逍遥散对慢性心理应激大鼠血清皮质酮及胃肠激素的影响

目的研究慢性应激状态下大鼠血清皮质酮(CORT)及胃肠激素(GAS、MOT)水平的变化及逍遥散与柴胡疏肝散的干预作用。方法大鼠分为空白对照组、模型组、柴胡疏肝散组及逍遥散组。除空白对照组外,各组均复制慢性多相性应激模型,同时,柴胡疏肝散组及逍遥散组分别灌服柴胡疏肝散及逍遥散21d,空白对照组与模型组灌服等容积生理盐水。以旷场实验观察各组大鼠的行为学变化,以放免法检测各组大鼠血清CORT、GAS及血浆MOT。结果与空白对照组比较,模型组大鼠体重明显降低(P<0.01),旷场活动次数明显减少(P<0.01),血清CORT明显升高(P<0.01),血清GAS及血浆MOT明显下降(P<0.01);与模型组比较,柴胡疏肝散组及逍遥散组大鼠体重明显增加(P<0.05,P<0.01),旷场活动次数明显增加(P<0.05,P<0.01),血清CORT明显下降(P<0.01),血清GAS与血浆MOT也明显回升(P<0.05)。结论慢性多相性应激通过对大鼠血清CORT、GAS及血浆MOT的影响,致使大鼠精神状态异常、胃肠功能紊乱以及生长发育减缓。中药柴胡疏肝散及逍遥散通过对大鼠CORT、GAS及MOT的调节来抑制慢性应激对大鼠精神状态、胃肠功能及生长状态的不良影响。

Effect of Chaihu Shugan Powder and Xiaoyao Powder on Serum Corticosterone and Gastrointestinal Hormones of Chronic Multi-stress Rats

Objective To observe the content changes of corticosterone and gastrointestinal hormones in serum of chronic multi-stress rats and to investigate the effect of Xiaoyao Powder and Chaihu Shugan Powder.Methods Wistar rats were randomly allocated to four groups:normal group(group A),model group(group B),Xiaoyao Powder group(group C),Chaihu Shugan Powder group(group D)etc.Chronic multi-sress models were established in all the rats except those of normal group.Xiaoyao Powder group and Chaihu Shugan Powder group were respectively given the corresponding drugs for 21 days.Normal group and model group were administered the equal amount of normal saline by gavage.After the administration,the open-field test was used to observe the behavior changes of the rats,and the radioimmunoassay method was adopted to detect the serum corticosterone and gastrin contents and plasma motilin level.Results Compared with the normal group,the body weight of model group was decreased(P < 0.01)and the open-field behavior frequency was reduced(P < 0.01),the corticosterone content in serum increased(P < 0.01),and gastrin in serum and motilin in plasma levels were decreased(P < 0.01).Compared with the model group,the body weight and open-field behavior frequency in Xiaoyao Powder group and Chaihu Shugan Powder group were increased(P < 0.05,P < 0.01),the open-field behavior frequency increased(P < 0.05,P < 0.01),the corticosterone content in serum decreased(P < 0.01),and the gastrin in serum and motilin in serocym levels increased(P < 0.05).Conclusions Chronic multi-stress can result in rats growth restraining,abnormal mental state and gastrointestinal dysfunction through increasing corticosterone and decreasing gastrin and motilin.Chaihu Shugan Powder and Xiaoyao Powder can restrain the influence of chronic multi-stress by regulating those hormones levels in rats body.
