人肺癌及癌旁组织p21蛋白表达的免疫组化检测

以相当于 ras p21蛋白一段氨基酸序列的合成肽的单克隆抗体 EB_(24?),用 ABC 免疫组化技术检测50例福尔马林固定-石腊包埋的人肺癌组织切片的 p21蛋白表达。46例癌组织的 p21表达,比癌旁病理形态证明“正常”的肺泡及正常肺泡明显增强.其中以肺腺癌最为显著。42例非小细胞肺癌中38例呈阳性反应(90.5%),强阳性约占60%;而8例小细胞肺癌中只有1例阳性(12.5%)。一般说肺腺癌对化疗最不敏咸.小细胞肺癌对化疗较为敏感。这种差别与 p21的表达是否有关联值得进一步探讨。

IMMUNOHISTOCHEMICAL DETECTION OF RAS p21 EXPRESSION IN HUMAN LUNG CARCINOMA AND ADJACENT NORMAL TISSUES

The expression of ras p21 was examined in 50 sections of formalin-fixed or paraffin- embeded human lung carcinoma and its adjacent normal tissues sing monoclonal antibody EB_(2411) to a synthetic peptide reflecting amino acid position 56-64 of ras p21 protein by immunohistoche- mical techniques.In 46 of 52 cases studied more p21 expression was observed in lung carcinoma tissues compared with their adjacent apparent normal tissues.The enhanced expression of p21 in adenocarcinoma of lung was more distinct than that in other types of lung carcinoma.Retivity of 38 of 42 non-small cell lung carcinoma tested with McAb EB_(2411)was positive(90.5%)and 60% of them were strong positive,while in small cell lung carcinoma(SCLC)tested only 1 of 8 cases appeared positive reaction.Generally,SCLC is more sensitive to chemotherapy than lung adenocarcinoma.It is worth to search after if this difference of chemotherapy sensitivity between various types of lung carcinoma is associated with ras p21 expression in tumor tissues.
