自然铜和鹿衔草对裸鼠肺癌骨转移的抑制作用

目的:探讨自然铜和鹿衔草对肺癌骨转移的抑制作用。方法:取15～20g裸鼠,每只裸鼠胫骨骨髓腔内注射0.1mL A549细胞悬液,14天后建立肺癌骨转移模型。随机分为中药组、阳性药物组、模型对照组。中药组按6g.kg-1隔日灌胃自然铜和鹿衔草,阳性药物组按10mg.kg-1每周腹腔注射帕米磷酸二钠,给药3周后处死,称量体重,测量肿瘤体积并计算抑瘤率,流式细胞仪检测肿瘤细胞凋亡率、血生化仪检测血钙、磷、碱性磷酸酶及肝肾功能等。结果:与模型对照组比较,中药组和阳性药物组的肿瘤体积缩小,血钙和碱性磷酸酶降低,肿瘤细胞凋亡率升高,尿素氮减少,而阳性药物组体重下降,天冬氨酸氨基转移酶升高;与阳性药物组比较,中药组体重增加,肿瘤体积缩小,抑瘤率提高,血钙、天冬氨酸氨基转移酶和丙氨酸氨基转移酶下降,尿素氮升高;3组血清磷和肌酐比较,差异无统计学意义。结论:自然铜和鹿衔草能安全有效地抑制肺癌骨转移导致的骨破坏。

Inhibitory Action of Pyrite and Pyrola on Nude Mouse with Bone Metastasis in Lung Cancer

Objective:To discuss the inhibitory action of pyrite and pyrola on bone metastasis in lung cancer.Methods:Take 15~20g nude mice,inject 0.1mL A549 cell suspension into marrow cavity of tibia of every nude mouse,after 14 days,establish the models of bone metastasis in lung cancer.Then they were randomly divided into Chinese medicine group,positive drug group and model control group.The Chinese medicine group were administered the pyrite and pyrola with 6 g kg-1 into stomach every other day.Positive drug group were injected into enterocoelia with pamidronate.Disodium 10 mg kg-1 every week.After three weeks,they were executed and weighed to measure the tumor volume and calculate the tumor-suppressing rate.Use the flow cytometer to detect apoptosis rate,blood biochemistry analyzer to calcium,phosphorus,alkaline phosphatase,hepatic/renal function and so on.Results:Compared to the model control group,in Chinese medicine group and positive drug group,the tumor volume has shrinked,calcium and alkaline phosphatase reduced,apoptosis rate increased and urea nitrogen decreased.And the weight of positive drug group descended,AST increased;by comparison,in Chinese medicine group,the weight elevated,the tumor bulk lessened,apoptosis rate increased,Ca,AST,ALT all fell down and the urea nitrogen has gone up;the differences of serum phosphorus and creatinine among the three groups have no statistical significance.Conclusion:Pyrite and pyrola can safely control bone destruction caused by bone metastasis in lung cancer.
