妊娠期糖尿病胎盘部位脂联素及其受体的表达

目的:探讨胎盘部位脂联素及其受体基因mRNA表达在妊娠期糖尿病病理状态下的变化趋势及与新生儿体重、新生儿Ponderal指数(PI)之间的关系。方法:对26例妊娠期糖尿病产妇、30例正常产妇采用逆转录聚合酶链反应(RT-PCR)技术检测胎盘脂联素及其受体基因mRNA的表达,并对其表达水平进行半定量分析。结果:①脂联素及其受体1/2基因mRNA表达于胎盘组织;②妊娠期糖尿病组胎盘脂联素基因mRNA表达水平(0.75±0.19)显著低于正常对照组(1.17±0.17),差异有统计学意义(P<0.05);妊娠期糖尿病组胎盘AdipoR1基因mRNA的表达水平(2.08±0.67)与正常对照组(1.98±0.68)比较,差异无统计学意义(P>0.05);妊娠期糖尿病组胎盘AdipoR2基因mRNA的表达水平(1.58±0.36)显著高于正常对照组(1.19±0.25),差异有统计学意义(P<0.05);③胎盘脂联素基因mRNA表达水平与新生儿出生体重、PI呈显著负相关关系;胎盘AdipoR2基因mRNA表达水平与新生儿出生体重、PI无相关性。结论:①胎盘组织中存在脂联素及其受体基因的表达;②胎盘来源的脂联素及其受体可能参与了胎儿生长发育的调节。

The study on the mRNA expression of adiponectin and its receptors in placenta of gestational diabetes mellitus

Objective:To investigate the variation of the mRNA expression of adiponectin and its receptors in placenta of gestational diabetes mellitus(GDM) and the relation between mRNA expressions of adiponectin and its receptors in placenta of GDM women and newborn's birth weight and Ponderal index(PI).Methods:The adiponectin and its receptors mRNA expression level in placenta of 26 mothers with GDM and 30 cases of normal controls were evaluated by RT-PCR.Results:①The adiponectin and its receptors mRNA were expressed in placenta.②The expression of adiponectin mRNA in GDM women's placenta(0.75±0.19) was significantly lower than that in the controls(1.17±0.17)(P<0.05);Between the expression of adiponectin receptor1 mRNA in GDM women's placenta(2.08±0.67) and in the controls(1.98±0.68) ,there were no significant difference (P>0.05);The expression of adiponectin receptor2 mRNA in GDM women's placenta(1.58±0.36) was significantly higher than that in the controls(1.19±0.25)(P<0.05).③The expression level of adiponectin mRNA in GDM mothers was negatively correlated with the birth weight and PI of their newborns; But the expression level of adiponectin receptor2 mRNA in GDM mothers was no correlated with the birth weight and PI of their newborns.Conclusion:①There are expression of adiponectin and its receptors mRNA in placenta.②Adiponectin and its receptors in placenta may participate in regulation of fetal development.
