摘    要:目的:观察基因修饰的骨髓基质细胞对辐射的耐受作用。方法:通过辐射对CFU-F形成的影响制作细胞存活曲线分析造血因子双顺反子基因修饰的基质细胞(HFCL/FG和HFCL/EFG)的对辐射的耐受,采用3H-TdR掺入法验证上述结果,观察转染细胞对粒系祖细胞生长的影响,结果:HFCL/FG和HFCL/EFG细胞生存曲线的肩明显增宽,D0值增大,N和Dq值增大,3H-TdR掺入实验也验证了该结果,这种细胞对粒系祖细胞的扩增作用明显增强。结论:造血因子基因修饰的骨髓基质细胞具有抗辐射和支持造血作用。

Abstract:Objective\ To explore radioresistance \|improving and radio\|protective effects of hematopoietic growth factor genes transfected into HFCL stromal cells. Methods\ Radioresistance of gene\|transfected cells to radiation was detected by dose survival curve and 3H\|TdR incorporation rate,Colony forming assay on cultured stroma layer was examined. Results\ The gene\|modified cells (HFCL/EFG and HFCL/FG) showed more resistant to ionizing radiation than control HFCL cells by dose survival curve, 3H\|TdR incorporation rate.The transfected genes had effects of protecting hematopoiesis and radioprotection by colony forming assay. Conclusion\ Stromal cells transduced with hematopoietic growth factor genes can improve radioprotective effects.
