电磁脉冲对大鼠肝脏线粒体膜流动性及脂质过氧化的影响

摘    要:目的 研究高场强电磁脉冲对线粒体膜的生物学效应。方法 大鼠肝脏线粒体悬液经6 0kV/m电磁脉冲作用 ,运用电子顺磁共振 (ESR)波谱技术检测线粒体膜蛋白及膜脂运动性的变化 ,并采用分光光度法检测膜脂质过氧化的变化。结果 膜蛋白运动性由旋转相关时间 (τc)表示 ,脉冲作用后为 (0 .5 94± 0 .0 4 9)× 10 -9s,对照组为 (0 .5 0 1± 0 .0 77)× 10 -9s,差异有显著性 (P <0 .0 5 ) ,膜蛋白运动变慢 ;线粒体膜脂的有序性由序参数 (S)表示 ,脉冲作用后为 0 .6 1± 0 .0 1,对照组为 0 .6 3± 0 .0 1,差异有显著性 (P <0 .0 5 ) ,膜脂的有序性降低 ,运动加快 ;脂质过氧化产物丙二醛 (MDA)的含量脉冲作用后有所降低。结论 电磁脉冲作用使线粒体膜运动紊乱 ,并且影响膜脂质的过氧化

Abstract:OBJECTIVE: To study the effects of intense electromagnetic pulse(EMP) on the biological effects of mitochondrial membrane. METHOD: Rat liver mitochondrial suspension was exposed to EMP at 60 kV/m level. The changes of membrane lipid fluidity and membrane protein mobility were detected by ESR and spin label technique. Malondialdehyde(MDA) was detected by spectrophotometer. RESULTS: The mobility of membrane protein decreased significantly(P < 0.05). Correlation time (tau c) of control group was (0.501 +/- 0.077) x 10(-9)s, and tau c of EMP group was (0.594 +/- 0.049) x 10(-9)s, indicating that the mobility of protein was restricted. The fluidity of mitochondrial membrane increased significantly(P < 0.05) at the same time. Order parameter(S) of mitochondrial membrane lipid in control group was 0.63 +/- 0.01, while S of EMP group was 0.61 +/- 0.01(P < 0.05). MDA decreased significantly. CONCLUSION: The mobility and lipid peroxidation of mitochondrial membrane may be disturbed after EMP exposure.
