摘    要:目的:建立测定人血清中阿莫西林浓度的RP—HPLC法。方法:采用甲醇沉淀蛋白,高效液相色谱检测法,以别嘌醇为内标,色谱柱为Phenomenex Synergi4μPOLAR-80A,250mm×4.6mm,流动相为甲醇-0.2%甲酸水溶液(含10mmol·L^-1乙酸铵)/4:96,流速1.2mL·min^-1,检测波长:236nm。结果:血清中内源性物质不干扰阿莫西林的检测,阿莫西林的线性范围为0.327~20.95/2g·mL^-1,批内、批间精密度均小于15%,低、中、高浓度的(0.655、2.619和10.475μg·mL^-1)平均提取回收率分别为102.9%、99.8%、97.6%。结论:该法操作快速、简单、准确,符合生物样品检测要求,可用于临床药代动力学研究中大批量血样的处理。

Abstract:OBJECTIVE: To develop a RP- HPLC method for determination of amoxicillin in human serum. METHODS: Serum protein was precipitated with methanol. With antigut used as internal standard, the analysis was conducted on Phenomenex Synergi 4 μ POLAR - 80A (250 mm×4.6 mm) column. The mobile phase consisted of Methanol - 0.2 % formic acid (containing 10 mmol·L^-1 ammonium acetate) at a flow rate of 1.2 mL·min^-1 and a detection wavelength of 236 nm. RESULTS: The detection was free of interference from serum endogenous substance. The linear concentration range of Amoxicillin was 0. 327 - 20.95 μg·mL^-1. The intra - and inter- day precision over the entire concentration range were less than 15%. The mean extraction recoveries of amoxicillin at concentrations of 0.655, 2.619 and 10.475μg·mL^-1 were 102.9%, 99.8% and 97.6%, respectively. CONCLUSION: The method is rapid, simple and accurate, meeting the standard for the detection of biological specimen, and it is applicable for the treatment of a large number of blood samples in the research of clinical pharmacokinetics.
