乌司他丁对百草枯急性中毒大鼠肺组织MMP-9mRNA表达的影响

摘    要:
目的 探讨乌司他丁(UTI)对百草枯(PQ)急性中毒大鼠肺组织基质金属蛋白酶-9(MMP-9)mRNA的影响及肺损伤的肺保护作用机制.方法 将80只SD大鼠随机分成8组,每组10只,分别为正常对照组、UTI组和PQ中毒后12、24、48h组及PQ+UTI 12、24、48h组,同时应用RT-PCR技术对各组大鼠肺组织中MMP-9的表达进行观察.结果 (1)通过大鼠行为学改变及肺组织切片证实PQ急性中毒模型成立.(2)PQ急性中毒组早期肺组织毛细血管扩张充血,肺组织炎症细胞浸润.(3) PQ组大鼠12、24、48h时肺组织MMP-9mRNA水平分别为(1.13士0.02)、(1.28士0.04)、(1.38士0.06),PQ组大鼠经UTI干预后上述各时段肺组织MMP-9mRNA水平分别为(1.05士0.03)、(1.09士0.05)、(1.12士0.05),差异均有统计学意义(均P<0.05).结论 UTI可明显减低PQ急性中毒大鼠肺组织MMP-9mRNA表达水平,可能是其防治PQ致肺损伤的机制之一.

The effect of unithiol on the expression of MMP-9 gene in the lung of paraquat poisoning rats

Abstract:
Objective To research effect of unithiol on the changes of MMP-9mRNA in the lung of paraquate poisoning rats. Methods Eighty male SD rats were divided into 8 groups (n=10) randomly:Normal control, UTI control ,PQ poisoning model (PQ groups, 12h,24h,48h groups), PQ+UTI model(PQ+UTI groups, 12h,24h,48h groups). PQ poisoning model were established by intraperitoneal injecting paraquate.The expression of MMP-9mRNA in lung tissues was detected by RT-PCR. Results (1)By observing the changes of action and observing the lung tissues sections,the authors demonstrated that the rats' PQ poisoning models was successfully established. (2)The histopathology of lung showed infiltration of inflammatory cell in acute phase(within 48hours), The Inflammation increased gradually within 48 hours, Howerer ,the changes of pathology was alleviated obviously in the UTI+PQ groups.(3) Compared with the PQ groups, the expression of MMP-9mRNA in lung diminished greatly in the PQ+UTI groups after being injected UTI(P〈0.05). Conclusion Down-regulation of the expression of MMP-9 by UTI may be one of mechanisms by which pulmonary injury are prevented in the acute paraquate poisoning.
