潮汕人载脂蛋白E基因多态性与2型糖尿病合并冠心病的相关性研究

摘    要:目的 探讨潮汕地区2型糖尿病(T2DM)患者载脂蛋白E(ApoE)基因遗传多态性与冠心病(CHD)的相关性.方法 采用实时荧光定量聚合酶链反应(RT-PCR)技术分别检测T2DM并发CHD组(T2DM+CHD)、T2DM组和健康体检组的ApoE基因型,并检测所有受试者血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HLD-C)、低密度脂蛋白胆固醇(LDL-C)和空腹血糖(FBG).结果 T2DM+CHD组的E4/E3基因型和E4等位基因频率均明显高于正常对照组和T2DM组(P<0.01).与携带E3等位基因的T2DM+CHD患者相比,携带E4等位基因T2DM+CHD患者体内TC、LDL-C浓度升高,两者之间的差异有统计学意义(P<0.05).结论 ApoE基因E4等位基因可能增高潮汕人T2DM患者并发CHD的遗传易感性.

Abstract:Objective To investigate the association of genetic polymorphisms of apolipoprotein E (ApoE) gene and type-2 diabetes mellitus (T2DM) complicated with coronary heart disease (CHD) in Chaoshan patients.Methods ApoE gene polymorphisms were detected by real-time PCR (RT-PCR) in the subjects of three groups (control group,T2DM group,and T2DM+CHD group).At the same time,triglyceride (TG),total cholesterol (TC),high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol (LDL-C),fasting blood-glucose (FBG) were detected in the serum of these subjects.Results Among the three groups,the frequencies of E4/E3 genotype and E4-allele of ApoE were the highest in T2DM+CHD group (P<0.05).The levels of serum TC and LDL-C of E4-allele carriers were higher than those of E3-allele carriers in T2DM+CHD group,with statistically significant differences (P<0.05).Conclusion ApoE E4-alleles increases the susceptibility to CHD in T2DM patients of Chaoshan region.
