流式细胞术检测冠心病患者外周血白细胞唾液酸黏附素的表达及临床意义

摘    要:目的观察唾液酸黏附素(Siglec-1)在冠心病(CHD)患者外周血淋巴细胞、单核细胞和中性粒细胞上的表达水平,并探讨其在冠状动脉粥样硬化发病过程中的作用。方法流式细胞术检测57例CHD患者及38名正常对照者外周血淋巴细胞、单核细胞和中性粒细胞Siglec-1的百分率;生化常规测定所有人选者血脂水平。结果流式细胞仪检测发现,Siglec-1在正常对照组及CHD组淋巴细胞和中性粒细胞上均无表达;CHD组单核细胞Siglec-1阳性率为(12.74-2.4)%,显著高于正常对照组[(1.0±0.3)%,P〈0.01],而CHD血脂正常组和血脂异常组的Siglec-1阳性率差异无统计学意义。结论作为单核巨噬细胞激活的标志,Siglec-1在外周血淋巴细胞和中性粒细胞上不表达,在正常人单核细胞上表达很低。CHD患者单核细胞Siglec-1表达明显升高,提示CHD患者外周血单核细胞已经激活,发生巨噬细胞化,Siglec-1在单核细胞黏附血管内皮及CHD的发病过程中可能起重要作用。

Abstract:Objective To investigate the expression of sialic acid-binding immunoglobulin-like lectin-1 (Siglec-1) on peripheral blood lymphocytes, monocytes and neutrophils in patients with coronary heart disease (CHD) and explore the role of Siglec-1 in coronary atherogenesis. Methods The percentage of Siglec-1 in lymphocytes, monocytes and neutrophils was determined by flow cytometry in 57 CHD patients and 38 healthy controls. The levels of serum lipids were determined by clinical chemistry analyzer. Results Siglec-1 protein was not found on lymphocytes and neutrophils in both CHD patients and healthy controls. The positive rate of Siglec-1 on monocytes in CHD patients was significantly higher than that in healthy controls[ ( 12.7 ± 2.4) % vs ( 1.0 ± 0.3 ) % , P 〈 0.01 ]. However, no significant difference was found between normal and abnormal levels of serum lipids in CHD patients. Conclusions As a major biomarker of the activation of monocytes/macrophages, Siglec-1 is not expressed on peripheral blood lymphocytes and neutro- phils and is lightly expressed on monocytes in healthy controls. The increasing expression of Siglec-1 in CHD patients demonstrates the activation of monocytes in peripheral blood. Siglec-1 may mediate monocytes adhere to vascular endothelial ceils and play an important role in the pathogenesis of CHD.
