摘 要: | ![]() 目的:研究脊髓损伤(SCI)后的不同时间点尾静脉移植大鼠骨髓间充质干细胞(BMSCs)对大鼠SCI的修复作用。方法:SD大鼠24只,制备大鼠SCI模型,随机分为3组各8只:对照组于SCI后尾静脉注射生理盐水;24 h移植组和于7 d移植组分别于SCI后24 h、7 d尾静脉注射BMSCs缓冲液(2×106/m L)1 m L。在SCI后各时间点分别进行Basso Beattie Bresnahan(BBB)运动学评分,于第42天处死动物,HE染色观察SCI处空洞面积的改变情况。结果:7 d移植组大鼠SCI后14、21、28、35、42 d的BBB评分明显高于24 h移植组和对照组,差异有统计学意义(P0.05)。HE染色显示各组SCI部位形成脊髓空洞,7 d移植组大鼠的脊髓空洞明显小于24 h移植组和对照组。结论:BMSCs可在体外培养、扩增,能通过尾静脉注射的方式迁移到SCI部位,促进SCI大鼠的神经功能恢复;移植的最佳时间在损伤后7 d左右。

Therapeutic Effect of Bone Marrow Mesenchymal Stem Cells on Spinal Cord Injury Rats |

Abstract: | ![]() Objective:To study the effect of rat bone marrow mesenchymal stem cells (BMSCs) on rats with spinal cord injury (SCI) at different time points. Methods: SCI models were established in 24 SD rats, and ran-domly divided into 3 groups of 8 rats in each: control group was injected with normal saline after SCI, groups 24 h transplantation and 7 d transplantation were injected BMSCs buffer (2×106/mL) 1 mL respectively at 24 h, 7 d after SCI through the tail vein. Basso, Beattie and Bresnahan (BBB) locomotor rating scale was used at different time points following damage. Rats were sacrificed at day 42, and cavition in the contused spinal cord were observe by HE staning. Results: The BBB scores of the 7 d transplantation group were better than those of the 24 h transplan-tation group and the control group at day 14, 21, 28, 35, 42 (P<0.05). HE staining showed that there were cavition in the contused spinal cord of 3 groups, and the cavition of the 7 d transplantation group was significantly less than the groups 24 h transplantation and control. Conclusion: BMSCs can be cultured and amplificated in vitro, can be injected through the tail vein to migrate to the SCI site, to promote the recovery of neurological function in SCI rats. And the best time is about 7 d after the injury. |
