摘 要: | 目的: 探讨银杏叶提取物可否拮抗同型半胱氨酸诱导的血管内皮细胞血凝素样氧化型低密度脂蛋白受体1 mRNA表达升高。方法:采用含不同浓度(0.001mmol/L、0.01 mmol/L和0.1 mmol/L)同型半胱氨酸的培养液孵育人脐静脉血管内皮细胞,观察同型半胱氨酸对血凝素样氧化型低密度脂蛋白受体1 mRNA的表达水平的影响;取最能诱导血凝素样氧化型低密度脂蛋白受体1表达升高的同型半胱氨酸浓度,分别加入银杏叶提取物(0.01mmol/L银杏苦内酯A,0.01mmol/L银杏苦内酯B和0.01mmol/L白果内酯)和B族维生素(0.01mmol/L 叶酸,0.01mmol/L维生素B6,0.01mmol/L维生素B12),观察银杏叶提取物对同型半胱氨酸诱导的血凝素样氧化型低密度脂蛋白受体1 mRNA表达升高的作用,并与B族维生素进行比较。结果:随着同型半胱氨酸浓度的增加,血凝素样氧化型低密度脂蛋白受体1 mRNA表达增加,0.1mmol/L 的同型半胱氨酸最能诱导血凝素样氧化型低密度脂蛋白受体1 mRNA的表达;同型半胱氨酸+银杏叶提取物组和同型半胱氨酸+B族维生素组血凝素样氧化型低密度脂蛋白受体1 mRNA表达较同型半胱氨酸组明显减少(吸光度值0.42±0.03和0.65±0.04 VS 0.83±0.05,P<0.05),同型半胱氨酸+银杏叶提取物组降低血凝素样氧化型低密度脂蛋白受体1 mRNA表达的幅度较同型半胱氨酸+B族维生素组更大(吸光度值0.42±0.03 VS 0.65±0.04,P<0.05)。结论:同型半胱氨酸具有诱导血管内皮细胞血凝素样氧化型低密度脂蛋白受体1 m RNA表达的作用,且呈剂量依赖性。银杏叶提取物可拮抗同型半胱氨酸诱导的血管内皮细胞血凝素样氧化型低密度脂蛋白受体1 mRNA表达升高,有望应用于治疗高同型半胱氨酸所致的动脉粥样硬化。

Antagonistic Effect of Ginbgo Biloba Extract on Letin-like Oxidized Low Density Lipoprotein Receptor-1 mRNA Expression Increasing Induced by Homocysteine |

Abstract: | Objective To investigate the effect of ginbgo biloba extract on letin-like oxidized low density lipoprotein receptor-1 (LOX-1) mRNA expression increasing induced by homocysteine. Methods LOX-1 mRNA expression was observed after Human umbilical vein endothelial cells having been cultured in medium containing different concentration of homocysteine. Ginbgo biloba extract (0.01 mmol/L ginkgolide A, 0.01 mmol/L ginkgolide B, 0.01 mmol/L bilogolide) and vitamin B(0.01 mmol/L folic acid, 0.01 mmol/L vitamin B6, 0.01 mmol/L vitamin B12) were added respectively with homocysteine whose concentration induced the highest expression of LOX-1 mRNA. Change of LOX-1 mRNA expression induced by ginbgo biloba extract was observed and compared with that induced by vitamin B. Results LOX-1 mRNA expression increased with the homocysteine concentration increasing. 0.01 mmol/L homocysteine induced the highest expression of LOX-1 mRNA. Considerable decreases of LOX-1 mRNA expression were observed in group homocysteine+ Ginbgo biloba extract and group homocysteine+ vitamin B. More decrease was observed in group homocysteine+ Ginbgo biloba extract compared with group homocysteine+ vitamin B. Conclusion Homocysteine increases the LOX-1 mRNA expression in dose dependent way. Ginbgo biloba extract can antagonize the LOX-1 mRNA expression increasing induced by homocysteine. It is promising for ginbgo biloba extract to be apply to atherosclerosis due to homocysteine. |
