红景天预处理对大鼠肝脏缺血再灌注损伤细胞凋亡的影响

摘    要:
目的:探讨胃饲红景天预处理对大鼠肝脏缺血再灌注损伤后细胞凋亡的影响及其对肝功能的保护作用。方法:48只健康成年雄性SD大鼠随机分为生理盐水对照组和红景天预处理组,两组分别予与等量生理盐水、红景天水煎剂(5g·kg^-1·d^-1生药材)灌胃5d,末次灌胃2h后经第一肝门阻断入肝血流30min而后松开无创动脉夹,恢复人肝血流,分别于缺血30min及再灌注后1h、6h及24h各时相点活杀动物取材,检测血清丙氨酸氨基转移酶(ALT)、肝组织丙二醛(MDA),并用TUNEL染色及图像分析法检测肝细胞凋亡率的变化。结果:与生理盐水对照组比较,红景天预处理组ALT、MDA、肝细胞凋亡率在上述多个时相点显著降低(P〈0.05)。结论:肝脏缺血再灌注前给予红景天水煎剂预处理可以显著降低肝细胞凋亡率,减少MDA含量,并能有效地保护肝脏功能。

Influence of Salidroside Gastric Feeding Pretreatment on Hepatocyte Apoptosis Induced by Ischemia-reperfusion Injury in Rats

Abstract:
Objective: To investigate the effects of salidroside gastric feeding pretreatment on hepatocyte apoptosis induced by ischemia-reperfusion injury, and to see if it has any protective effect to liver function. Methods: Forty-eight healthy male adult SD rats were divided randomly into saline control group (group A, n = 24), and salidroside pretreatment group (group B, n = 24) and pretreated with saline or salidroside (5g·kg^-1·d^-1) by gastric feeding respectively for five days. Liver blood flow was obstructed for 30 min by clamping the first porta hepatic in 2 h after the last administration and restored when bulldog clamp was removed. Animals were sacrificed immediately after the ischemia and in 1 h, 6 h, and 24 h after the reperfusion respectively. Their serum alanine aminotransferase (ALT) and liver malonaldehyde (MDA) were determined. Apoptosis in liver was observed with TUNEL (TdT-mediated dUTP nick end labeling) stained sections, and analyzed with image analysis technique. Results: ALT serum levels and liver tissue MDA concentrations, and hepatocyte apoptosis index were markedly lower in group B than those in group A (P 〈 0.05 ) at many time points after liver reperfusion. Conclusion: Pretreatment with salidroside by gastric feeding in rats before ischemia-repcrfusion could obviously decrease hepatocyte apoptosis, reduce lipid peroxidation, and protect liver function effectively.
