白藜芦醇对人肾癌细胞增殖和细胞周期的影响

摘    要:目的探讨白藜芦醇(Res)对人肾癌细胞786-0(以下简称肾癌细胞)增殖和周期的影响。方法分别采用12.5、25、50、100μmol/L的白藜芦醇(Res)作用于肾癌细胞24 h;并设空白对照组。流式细胞仪检测细胞细胞周期。结果各浓度Res对肾癌细胞增殖均具有明显的抑制作用,表现为G1期及G2期细胞比例明显降低,S期比例明显升高,与对照组比较,P均〈0.05。Res 12.5μmol/L与25、50及100μmol/L比较,P均〈0.05;但25μmol/L与50、100μmol/L比较,P〉0.05。12.5μmol/L的Res抑制作用明显,当浓度达到25μmol/L时,抑制作用达最强;而继续升高浓度不能提高抑制作用。结论 Res对人肾癌细胞增殖有抑制作用,且具有剂量依赖性。

Abstract:Objective To investigate the influence of resveratrol(Res) on the proliferation and cell cycle of 786-0 human kidney cancer cells.Methods The Res of 12.5,25,50,100 μmol/L were used to treat kidney cancer cells for 24 h;blank control group was set.The cell cycle phase distribution of kidney cancer cells were analyzed by using flow cytometry.Results Res of all concentrations inhibited the proliferation of kidney cancer cells,the ratio of cells in G1/G2 phase were decreased conspicuously and the S phase were apparently increased,the difference has statistics significance compared with the control group(P0.05).The Res groups compared with each other,the 12.5 μmol /L group with 25 μmol/L group,50 μmol/L group,100 μmol/L group had significant difference(P0.05);but the 25 μmol/L group and 50 μmol/L group,100 μmol/L group had no significant difference(P0.05).Conclusions Human renal cancer cells can be inhibited in a dose-dependent by Res,and it can be visibly inhibited by the 12.5 μmol/L of Res.The inhibitory affect on human renal carcinoma cells proliferation reach peak when the concentration of Res hits 25 μmol/L,and it cannot be improved when continue to rise the concentration of Res.
