摘    要:目的:研究糖尿病大鼠膀胱细胞凋亡情况及其意义.方法:实验分为正常对照组和糖尿病组.用10周龄SD大鼠腹腔内注射1%链脲佐菌素(STZ)建立糖尿病大鼠的动物模型.12周后取两组大鼠膀胱组织标本,原位细胞检测法检测细胞凋亡;免疫荧光方法检测CD146、PDGF-Rβ、NG-2表达;Western Blotting检测CD146、PDGF-Rβ表达.结果:与正常对照组比较,糖尿病组大鼠血糖升高,体质量减轻,膀胱湿重增加;细胞凋亡主要发生在糖尿病大鼠膀胱的尿路上皮细胞,而正常对照组未见明显尿路上皮细胞凋亡;两组大鼠膀胱组织中未见明显NG-2表达,CD146阳性表达细胞位于膀胱黏膜下层的血管壁及膀胱逼尿肌肌膜,PDGF-Rβ阳性表达细胞位于膀胱逼尿肌之间血管壁细胞及黏膜下血管壁;Western Blotting 检测结果显示,两组大鼠膀胱壁组织CD146、PDGF-Rβ的蛋白表达水平比较,差异无统计学意义(P>0.05).结论:糖尿病大鼠膀胱上皮细胞早期发即生凋亡改变,而周细胞早期凋亡不明显;上皮细胞的凋亡可能是糖尿病膀胱的治疗靶标之一.

Abstract:Objective: To investigate the cell apoptosis and its clinical significance in urinary bladder of diabetic rats. Methods: All rats were assigned to the normal control group and diabetic cystopathy group. Diabetic cystopathy models were established in 10-week SD rats by intraperitoneal injection of 1% streptozotoein. At week 12, the bladder tissues were harvested and the cell apoptosis in the tissue was detected with in-situ assays. The expression of CD146, PDGF-R~ and NG-2 was detected by immunofluorescence assay and expression of CD146 AND PDGF-RI3 by Western Blotting. Results: Compared with normal control group, diabetic cystopathy group showed high blood glucose, lowered body weights and increased wet-weight of the urinary bladder. Apoptosis of urothelial cells was found in diabetic cystopathy group, but not in NC group. Expression of NG-2 was negative in both groups. In addition, CD146-expressing cells were detected on vascular walls and the detrusor membrane in the cystic submucosa. By contrast, PDGF-R 3positive expression was found in the cells lining vascular walls between beth detrusors as well as in submocosal vascular walls. No signi~eant difference ( P 〉 O. 05) was revealed in the expression of CD146 and PDGF-R3 between both groups, as detected by Western blotting. Conclusion: Apoptosis of urothelial cells, but not pericytes, is involved in early-stage diabetic cystopathy in rats. Urothelial apoptosis may become one of the targets for treatment of diabetic cvstoDathv.
