Suppressive effect of low-dose SOM230 on bile duct ligation induced liver fibrosis in rats

Objective To investigate the effect of long-acting somatostatin analog SOM230 in rat models of bile duct ligation-induced liver fibrosis.Methods Eighty male Spratue-Dawley rats were randomly divided into five groups as follows：bile duct ligation,high dose SOM230（80mg/Kg）,low dose SOM230（8 mg/kg）.sham operation and normal control.Biochemical markers in serum,hydroxyproline and histology were determined on 2 weeks and 4 weeks after operation,respectively.Results In histopathological examination,low dose of SOM230 improved bile duct proliferation and collagen deposition,which could be demonstrated by semiquantitative grades of liver fibrosis.At 2 weeks（1.50±0.45 vs.2.08±0.38） and 4 weeks（2.5±0.32 uv.3.17±0.26） after operation,grade in low dose group was lower compared with model group.Low dose of SOM230 decreased the level of hydroxyproline compared with model group at 2 weeks（112.5±21.5 vs.167.8±23.4）evidently.High dose of SOM230 did not improve fibrosis which may be associated with the induction of high y-GT,ALP and TB by high dose of SOM230.Neither high dose nor lowdose of SOM230 could significantly improve the liver function and cholestasis,and affect the serum albumin.Conclusion Low dose of SOM230 can reduce collagen deposition in the liver,but neither high dose SOM230 nor low dose SOM230 injected subcutaneously improve liver function.
