CETP基因多态性与维吾尔族2型糖尿病关系

摘    要:
目的 探讨胆固醇酯转运蛋白(cholesteryl ester transfer protein,CETP)基因TaqⅠ酶切多态性与维吾尔族2型糖尿病(T2DM)及其体内脂质谱的关系.方法 依据病例对照设计方法,采用全自动生化分析仪等技术测地尔族体内脂质谱;应用聚合酶链反应-限制性内切酶长度多态性(PCR-RFLP)方法,对维吾尔族的53例T2DM患者、53例糖耐量异常患者和106名糖耐量正常者的CETP基因TaqⅠ酶切多态性进行比较分析.结果 3组人群空腹血糖(FPG),正常组为(5.14±0.24)mmol/L,T2DM组为(10.79±3.34)mmol/L,糖耐量异常组为(5.97±0.41)mmol/L(F=205.87,P=0.000);CETP基因型在3组人群中的分布差异有统计学意义(P=0.028);T2DM组、正常组、异常组的H+H-基因型频率分别为69.8%,45.3%,41.5%;T2DM组明显高于其他2组;不同基因型的空腹血糖(FPG),H-H-为(6.04±1.72)mmol/L,H+H+为(6.03±1.63)mmol/L,H+H-为(7.48±3.60)mmol/L,差异有统计学意义(P=0.001).对T2DM组和正常组进行Logistic回归分析显示,T2DM的独立危险因素依次是甘油三酯(TG)(P=0.000)、CETP基因型(P=0.007)和腰围(P=0.048),H+H-基因型患T2DN的危险性是H-H-的4.042倍(95%CI=1.243~13.149,P=0.020).结论 CETP基因TaqⅠ酶切多态性与新疆维吾尔族T2DM的危险性密切相关,H+H-基因型可能是T2DM的遗传危险因素.

Abstract:
Objective To study the association of TaqⅠ gene polymorphism with type 2 diabetes and lipodogramme in Uygur people.Methods On basis of case control design,lipodo gramme of Uygur people was determined by automatic biochemical analyzer technology and the polymorphism of cholesteryle stertransfer prote in(CETP)was analyzed by PCRR FLP in 53 type 2 diabetes mellitus(T-DM)patients,53 patients with impaired glucose to lerance(IGT)and 106 normal control of Uygur nationality.Results The fasting plasma glucose(FPG)in normal group was 5.14±0.24 mmol/L,10.79±3.34 mmol/L in T-DM group,and 5.97±0.41mmol/L in IGT group(F=205.87,P=0.000),respectively.CETP genotype distribution was significant different between the three groups(P=0.028).The frequency of H+H-geno type for T-DM was significantly higher than that of others(69.8%,45.3%,41.5% for TDM,IG T,and normal group,respectively).The values of FPG was 6.04±1.72 mmol/L for subjects with H-H-genotype,6.03±1.63 mmol/L with H+H+,and 7.48±3.60 mmol/L with H+H,respectively,with significan difference between the groups(P=0.001).The results of logistic regression showed that the dependent risk factors of T-DM were total glucose(P=0.000),CETP genotype(P=0.007),waist circum ference(P=0.048).The subjects with H+H- had a 4042 folds(95% CI=1.243-13.149,P=0.020)risk of T-DM compared with subjects with H+ H- genotype.Conclusion CET Pgene TaqⅠ polymorphism is closely related to the risk of T-DM and the H+H genotype may serve as a genetic risk factor of T-DM among Uygur people.
