C-MYC癌基因与消化系统肿瘤关系的研究

摘    要:目的:研究消化系统肿瘤中的C-MYC基因扩增的情况。方法:用聚合酶链式反应(PCR)测定84例老年患者恶性肿瘤中的C-MYC基因,通过激光密度扫描仪对EB染色后的琼脂糖凝胶进行积分光密度分析。结果:正常组织无C-MYC扩增,C-MYC/N+L-MYC比值95%可信区间为0.0869~0.6257,胃癌组织C-MYC基因扩增阳性率为69.5%,直肠癌组织的C-MYC基因扩增阳性率为62.2%,肝癌组织C-MYC基因扩增阳性率为55.6%,145例肿瘤组织C-MYC扩增总阳性率为65.5%。结论:癌组织C-MYC扩增高于正常组织,在不同消化系统肿瘤组织中扩增程度不同,与细胞潜在的增殖能力成正相关,对消化系统肿瘤的发生、发展及预后,特别是指导临床基因治疗提供有用的资料。

Abstract:Objective: To study the amplification of C-MYC gene in gastroenterological carcinoma.Methods:145 cases with different tumors were detected by polymerase chain reaction (PCR), and the results staining with EB were analysised by integrating density.Results:The amplification of C-MYC gene was not found in 32 normal controls, and the average density of C-MYC/N+L-MYC was 0.086 9~0.625 7. The positive rate of C-MYC amplification in 45 colon carcinoma was 62.2%, the positive rate of C-MYC amplification in 82 stomach cancers renal was 69.5%, the positive rate of C-MYC amplification in 18 hepatomas renal was 55.6%, the positive rate was 65.5% in the total 145 cases with different cancers.Conclusion:C-MYC gene was related with cell growth, differentiation and cancer, the detecting C-MYC amplification was useful for gene therapy with antisense oligonucleotide to C-MYC.
