不同光动力剂量对C6胶质瘤细胞胞内游离钙的影响

摘    要:目的 研究不同剂量光动力作用后C6胶质瘤细胞内游离钙[Ca^2 ]i的变化,探讨钙超载在光动力诱导C6胶质瘤细胞坏死和凋亡过程中的作用。方法 常规培养C6胶质瘤细胞,以不同光动力剂量处理对数生长期的实验组细胞后,应用Fluo-3/AM为细胞内钙离子荧光指示剂,激光共聚焦显微镜测定各组C6胶质瘤细胞内游离钙的变化;MTT法检测各组细胞的死亡率。结果 光动力作用后C6胶质瘤细胞内游离钙明显升高,平均胞内钙离子浓度和细胞死亡率随着光动力剂量增加而增大。结论 光动力作用后C6细胞钙超载在光动力诱导肿瘤细胞坏死和凋亡过程中起重要作用。

Abstract:Objective To investigate the change inCa 2+ i in C6glioma cell after PDT with diff erent doses and discuss the importan t role of calcium overload in PDT induced C6glioma cell necrosis and apoptosis.Methods The C6gliomas cells were grown in RPM I 1640medium supplemented with 10%f etal calf serum,then the cells were h arvested during the log phase of growth and tre ated by hematoporphyrin monomethyl ether(HMME)-mediated photodynamic therapy wit h different doses,respectively.The change inCa 2+ i in C6glioma cell following PDT was detected by means of a confocal laser s canning microscope using the Ca 2+ indicator Fluo -3/AM.The mortality of C6glioma cell under different out put and time of laser were measured by means of MTT reduction assay.Results The mortality of C6cell and the concentration of free Ca 2+ increased with the increase in light dosage during the early stage of PDT,correlated with energy density of th e irradiation.Conclusions The calcium overload may play an important role in PDT induced cell death r esponse processes.
