细胞凋亡和相关基因Fas、bax和bcl-2蛋白在睾丸肿瘤中表达的研究

摘    要:目的研究细胞凋亡和相关基因Fas、hax和bcl-2蛋白在睾丸肿瘤发生和发展中的作用。方法应用DNA未端标记、流式细胞术和细胞免疫荧光技术检测56例睾丸肿瘤细胞凋亡状态及凋亡相关基因Fas、bax和bcl-2蛋白的表达。结果凋亡指数与肿瘤病理类型和淋巴结转移密切相关(P<0.05)。Fas蛋白阳性表达37例(66.1%),bax阳性48例(85.7%),bcl-2阳性40例(71.4%);Fas、bax和bcl-2蛋白表达与肿瘤的病理类型、临床分期和淋巴结转移无明显相关(P>0.05):凋亡指数在Fas、bax及bcl-2蛋白阳性表达和阴性表达者中差异有显著性(P<0.05)。结论细胞凋亡在睾丸肿瘤发生中有重要作用。Fas、bax和bcl-2蛋白的异常表达尚不能成为睾丸肿瘤发生和发展的预后参数。

Abstract:Objective To explore the role of apoptosis and Fas, bax and bcl-2 proteins in the pathogenesis of testicular carcinoma. Methods Cell apoptosis was determined with TUNEL and flow cytometry, and the expression of Fas, bax and bcl-2 proteins were detected by cellular immunofluoresence technique in 56 cases of testicular carcinomas. Results Apoptotic index was closely correlated with the pathological type and lymph node metastasis of testicular carcinomas(P<0.05). The positive rates of Fas, bax and bcl-2 proteins expression in testicular carcinoma were 66.1% (37/56), 85.7% (48/56) and 71.4% (40/56), respectively. Fas, bax and bcl-2 proteins expression was not correlated with the pathological type, clinical stage and lymph node metastasis of testicular carcinomas (P>0.05). The apoptotic index was positively correlated with Fas and bax expression, and negatively correlated with bcl-2 expression (P<0.05). Conclusion Cell apoptosis may play a role in the pathogenesis of testicular carcinoma. Fas, bax and bcl-2 protein overexpression could not serve as a marker for the prognosis of testicular carcinoma.
