电针对坐骨神经损伤模型大鼠行为学及脊髓中TrkC表达的影响

摘    要:目的:观察电针对坐骨神经损伤大鼠行为学、神经生长因子受体TrkC的影响,探讨电针治疗坐骨神经损伤的生物学机制。方法:采用神经夹持损伤的方式建立大鼠坐骨神经损伤模型,通过SFI指数、BBB评分观察正常组、假手术组、模型组、模型对照组与电针组大鼠的行为学变化;通过免疫组织化学染色观察各组大鼠脊髓内TrkC表达情况,进而统计分析各组间的差异。结果:(1)模型组、模型对照组、电针组大鼠SFI指数与正常组相比明显降低(P<0.05),电针组与模型组相比,无统计学意义(P>0.05);(2)模型组、模型对照组、电针组大鼠BBB评分与正常组相比明显降低(P<0.05),电针组与模型组相比明显升高(P<0.05),但与正常组相比仍有显著性差异(P<0.05);(3)模型组和模型对照组TrkC平均光密度与正常组相比明显升高(P<0.05),电针组与模型组相比明显升高(P<0.05)。结论:电针治疗可以通过提高神经生长因子高亲和力受体TrkC的释放,促进神经元生长、分化和成熟,最终改善坐骨神经损伤大鼠的运动功能。

Abstract:Objective : To observe the influence of electro - acupuncture on the ethology and TrkC expression. And to investigate the biological mechanisms of electro - acupuncture in sciatic nerve injury. Methods : Sciatic nerve clamping injury was used as the model. The SFI and BBB tests were operated to observe the behavior changes. The level of TrkC in the spinal cord was measured by immunohistochemistry. Results : ( 1 ) SFI : Compared with the blank control group, that of model group, model control group and electro - acupuncture group decreased significantly (P 〈 0.05). The difference between the electro - acupuncture group and the model group was not statistically significant(P 〉 0. 05). (2)BBB: Compared with the blank control group, that of model group, model control group and electro - acupuncture group decreased significantly ( P 〈 0. 05 ). The BBB score of electro - acupuncture group was significantly higher than that of model group (P 〈 0. 05 ). The difference between the electro - acuouncture grouo andblank control group was statistically significant ( P 〈 0. 05 ). ( 3 ) TrkC : The AODs of TrkC in model group and model control group were significantly higher than that in blank control group ( P 〈 0.05). The AODs of TrkC in electro - acupuncture group were sig- nificantly higher than that in model group (P 〈 0. 05 ). Conclusion: Electro -acupuncture could treat sciatic nerve injury by in- creasing the release of TrkC. As a result,the motor function of the sciatic nerve injured rats is improved.
