摘    要:
目的探讨RHBDD1和EGFR在三阴型乳腺癌(triple negative breast cancer, TNBC)发生、转移中的作用及其临床意义。方法选取30例TNBC(ER、PR、HER2均阴性)和30例非三阴型乳腺癌(non triple negative breast cancer, NTNBC)(ER、PR、HER2至少一项阳性)患者作为研究对象,采用免疫组化、酶联免疫法和qRT-PCR技术检测组织和血清中RHBDD1和EGFR mRNA含量及蛋白表达。结果 TNBC组织中RHBDD1和EGFR蛋白阳性率(83%、80%)高于NTNBC组织(67%、37%),差异均有统计学意义(P<0.05);TNBC患者血清中RHBDD1和EGFR的浓度含量亦明显高于NTNBC患者,差异均有统计学意义(P<0.05);TNBC组织中的RHBDD1 mRNA和EGFR mRNA含量分别是癌旁组织的3.68倍和4.56倍,差异均有统计学意义(P<0.05)。RHBDD1和EGFR的表达与TNBC患者年龄无关,与临床分期、组织学分级、肿瘤大小、淋巴结转移以及远处转移呈正相关。...

Abstract:
Purpose To investigate the role and clinical significance of RHBDD1 and EGFR in the occurrence and metastasis of triple negative breast cancer(TNBC).Methods Thirty cases of triple negative breast cancer(ER,PR and HER2 were negative)and 30 cases of non triple negative breast cancer(NTNBC)(ER,PR or HER2 were at least one positive)were selected as the study subjects.The mRNA and protein expression of RHBDD1 and EGFR were detected by immunohistochemistry,enzyme-linked immunoassay and qRT-PCR.Results The positive rate of RHBDD1 and EGFR in TNBC(83%,80%)was higher than that in NTNBC(67%,37%).The difference was statistically significant(P<0.05).The serum levels of RHBDD1 and EGFR in TNBC patients were significantly higher than that of NTNBC(P<0.05).The levels of RHBDD1 mRNA and EGFR mRNA in TNBC tissues were 3.68 times and 4.56 times as much as those in adjacent cancer tissues,Their difference was statistically significant(P<0.05).The expression of RHBDD1 and EGFR in TNBC was positively correlated with clinical stage,histological grade,tumor size,lymph node metastasis and distant metastasis except age.There was a positive correlation between the expression of RHBDD1 and EGFR in TNBC(r=3.723).Conclusion The high expression of RHBDD1 and EGFR in TNBC,especially in lymph nodes and distant metastasis group suggests that RHBDD1 and EGFR play an important role in the occurrence and metastasis of TNBC,which can be used as TNBC molecular targets and provide theoretical guidance basis for clinical treatment.
