十二井穴流注放血干预急性脑缺血模型大鼠HSP70及CD31表达的研究

摘    要:目的探究十二井穴流注放血对急性脑缺血(ACI)模型大鼠HSP70及CD31表达的作用。方法60只SD大鼠随机分为假手术组、模型组、普通放血组、流注放血1组、流注放血2组、西药组,每个组10只,局灶性凝结大脑中动脉法制备ACI模型后分组治疗。神经功能缺陷评分量表(m NSS)评价治疗前后各组大鼠神经功能,HE染色观察各组大鼠脑组织病理变化。免疫组织化学观察大鼠缺血区脑组织内HSP70及CD31蛋白表达变化。结果各组大鼠治疗后m NSS评分均较治疗前显著降低,西药组改善大鼠mNSS评分效果优于普通放血组和流注放血1组并与流注放血2组治疗效果无显著性差异。HE染色结果显示流注放血2组及西药组大鼠脑组织病理结构较模型组显著恢复,神经元形态完整。免疫组化结果显示,治疗后各组大鼠脑组织中HSP70及CD31蛋白表达均高于模型组(P<0.05),西药组HSP70及CD31含量显著高于普通放血组和流注放血1组(P<0.05),并与流注放血2组无明显差异(P>0.05)。结论十二井穴流注放血可能通过促进HSP70及CD31的表达发挥对ACI大鼠的保护作用。

Abstract:Objective:To investigate the effects of Flowed-insertion Bloodletting at Twelve Jing-well Acupoints(FBTJA)on HSP70 and CD31 in rats with acute cerebral ischemia(ACI).Methods:A total of 60 SD rats were randomly divided into 6 groups:sham operation group,model group,normal insertion bloodletting group,FBTJA-1 group,FBTJA-2 group and positive control group.Middle cerebral artery obstruction was used to copy ACI model.The neurological function scores were measured by modified-Neurological severity scales(m NSS).HE staining was used to observe the pathological changes in the brain tissue.Immunohistochemistry(IHC)was used to detect the expression and distribution of HSP70 and CD31 in brain tissues.Results:The mNSS scores after treatment of rats in each group were significantly lower than before.The positive control group had the lowest mNSS score and it showed no difference with FBTJA-2 group.HE staining showed that the pathological structure of brain tissue in FBTJA-2 and positive control group was significantly restored when compared with the model group,and the neuron morphology was intact.IHC showed that the expressions of HSP70 and CD31 in all the treatment group were both higher than in the model group.The contents of HSP70 and CD31 in the positive control group were significantly higher than in normal insertion bloodletting group and FBTJA-1 group,while there was no difference between the positive control group and FBTJA-2 group.Conclusion:The Flowed-insertion Bloodletting at Twelve Jing-well Acupoints may alert the protective effects on rats with ACI by promoting HSP70 and CD31 expression.
