超氧阴离子所致的红细胞膜氧化损伤模型

摘    要:用邻苯三酚在碱性条件下自氧化产生的超氧阴离子(O_2~-)处理人红细胞膜导致细胞膜过氧化损伤,表现为膜流动性下降(P<0.01);膜荧光物质含量增加;出现由膜蛋白交联形成的高分子聚合物(HMP)以及膜重新封闭能力丧失。而外加超氧歧化酶后,可抑制膜氧化损伤作用的发生。上述结果表明,由O_~-2所致的人红细胞膜过氧化损伤可作为一种实验模型,用于探讨自由基(如O_2~-)损伤的分子机理以及验证、筛选某些抗衰老药物。

Abstract:The treatment of human erythrocyte membranes with superoxide anion ( 02 )produced by pyrogallic acid, pH 8.35, does peroxidative damages to cell membranes, such as the decrease of the membrane fluidity (P<0.01), the increase of the fluorescent substance on the membranes, the formation of high molecular polymers (HMP) due to the membrane protein crosslinking and the loss of ability of ghost's reseal. These damages, however, are not almost induced after adding superoxide dism-utase. It is considered that the peroxidative damage of 02 produced by pyrogallic acid to human erythrocyte membranes can act as a model of research on the molecular mechanism of peroxidative damage of free radicals such as 02 and also a model of verification and screening of some aatisenile drugs.
