鼻咽癌变过程中基因表达的cDNA阵列研究

摘    要:目的 :比较研究鼻咽癌变过程中不同阶段鼻咽上皮组织基因表达谱的差异 ,观察鼻咽癌发生中多个基因的作用 ,寻找与鼻咽癌变相关的基因。方法 :用二亚硝基哌嗪 (N ,N’Dinitrosopiperazine ,DNP)诱导大鼠鼻咽癌 ,获取癌变过程中单纯增生、异型增生和癌组织 ,提取总RNA后逆转录标记成cDNA探针 ,与 5 88个基因的AtlasTM RatcDNAExpressionArray杂交 ,灰度扫描杂交信号及统计分析差异。结果 :在鼻咽癌组织中 ,PDGFB ,M6 P IGFR2 ,ErbB3EGF等瘤基因抑瘤基因、CCNE ,CCND3等细胞周期调节基因以及NF κB ,JNK转录因子和细胞信号转导等基因表达升高。鼻咽异型增生组织表达升高的基因较癌组织少 ,有prothymosin alpha,Cu ZnSOD1 ,MAPK1 等基因表达升高 ,但较鼻咽单纯性增生组织高表达基因多 ;鼻咽上皮组织单纯性增生仅有个别基因表达升高 ,大部分基因表达与正常鼻咽组织基因表达无明显差异。结论 :多个基因的表达变化在鼻咽癌变过程中起作用 ,随癌变进程基因逐步高表达

Abstract:Objective To study the gene expression profile of nasopharyngeal tissues, and to observe the change of gene expression in tumorigenesis stages. Methods Rat nasopharyngeal carcinomas were induced with N,N’Dinitrosopiperazine (DNP). Simple hyperplasia, atypical hyperplasia, and cancer nasopharyngeal tissues were obtained, from which the total RNAs were respectively extracted. Reverse transcription probes from the RNAs were respectively hybridized to the Atlas TM Rat cDNA Expression Array. Results Most of the genes in the array highly expressed, such as PDGFB, M 6P/IGFR 2, ErbB 3EGF, CCNE, CCND 2, NF κB and JNK, and distinctly increased compared with normal nasopharyngeal tissues. Several genes in atypical hyerplasia, such as prothymosin alpha and Cu Zn SOD, were up regulated compared with normal tissues. Othre genes expressed as normal tissues. Conclusion More than one gene contributed to nasopharyngeal tumorigenesis, the expression of which increased along with tumorigenesis.
