摘    要:
目的:观察大鼠骨髓基质细胞在体外培养的条件及生物学特性并加以鉴定.方法:取Wistar大鼠骨髓,分离培养骨髓基质细胞(BMSC),相差显微镜下观察其形态,传代后观察其生长特性,用免疫组化方法加以鉴定.结果:原代培养20d后可分离得到BMSC,在5代以前生长形态相对稳定,典型的BMSC可分为两个类型.结论:BMSC具有贴壁生长特性,较易对其进行分离扩增,增殖速度快,可用于多种疾病的细胞治疗.

Abstract:
Objective: To observe in vitro culturing con dition and biological characteristics of bone marrow stromal cells(BMSC) of Wist ar rats and identify the cells. Methods: We separated bone mar row of the Wistar rats and cultured the BMSCs. The morphology of the cells was s tudied with a phase contrast microscope and the living characteristics after su bculture of the cells were observed. We identified the cells by using immunohist ochemical assessment. Results: BMSC could be separated 20 days after primary culture. The living characteristics of the cultured cells were qu ite stable before the 5th generation. Typical BMSC could be divided into 2 types . Conclusion: BMSC, which can adhere the wall, can be easily s eparated and quickly proliferated by culture, and can be used in cell therapy in many kinds of diseases.
