甲状腺功能低下对大鼠中枢神经细胞增殖及其迁移的影响

摘    要:
妊娠大鼠用丙基硫氧嘧啶造成甲状腺功能低下(甲低)后,其新生仔鼠~3H-胸腺嘧啶核苷示踪,在注射后不同时间取脑制成切片并进行自显影。结果表明:甲低时,①嗅球嗅室壁室管膜及其下层细胞增殖延迟,细胞向内颗粒层迁移的速度减慢;②齿状回多形层细胞向颗粒层迁移的速度减慢;③小脑皮质外颗粒层细胞增殖延迟,外颗粒层细胞向内颗粒层迁移的速度减慢;④大脑皮质视、听区侧脑室壁室管膜及其下层细胞向浅层迁移的速度减慢,致使浅层的形成延迟。作者认为细胞增殖及迁移的延缓,影响神经回路的建成,可能是克汀病患者聋哑、智力低下、步态不稳的原因之一。

Abstract:
Impacts of hypothyroidism on cell proliferation and migration in the developing rat brain were studied by autoradiograpby.Results showed that hypothyroidism could reduce proliferation cells, prolong proliferating time in the developing brain; retard the cell migration from ependymal and subependymal layers to internal granule layer in olfactory bulb; delay the migration from the polymorphocyte layer to granular layer in dentate gyrus; decrease the migration from the external gronular to internal granular layer in cerebellum; lower the migration rate from ependymal and subependymal layers to superficial layers of optic and auditory cortex in cerebrum.The retardation of cell proliferation and migration may disturb the establishment of neural synaptic connection and neural circuits. These may be one of the causes of clinical manifestation in cretinism.
