小柴胡汤对体外培养人肝癌细胞株HepG2增殖的影响

摘    要:目的探讨小柴胡汤45%乙醇提取物对体外培养人肝癌细胞株HepG2增殖的影响及其可能的机理。方法采用ATP-Lite法测定不同浓度小柴胡汤对体外培养的HepG2增殖的作用,同时利用氧自由基清除能(ORAC)法测定样品的抗氧化活性;以LPS刺激小鼠巨噬细胞RAW264.7产生一氧化氮(NO),Griess法测定NO的终产物亚硝酸盐的含量,观察样品对NO生成的影响。结果小柴胡汤在62.5~500mg/L浓度范围内可以显著抑制HepG2增殖,抑制作用呈剂量相关性,计算半数抑制浓度(IC50)为(90.7±23.7)mg/L。小柴胡汤提取物具有一定的抗氧化活性,其抗氧化能力为阳性对照维生素C的1.3倍。小柴胡汤在3.125~50mg/L浓度范围内均可以显著抑制NO的生成(P0.05,P0.01)。结论小柴胡汤具有体外抑制人肝癌细胞株HepG2增殖的作用,其抑制作用可能与其抗氧化活性及抑制NO生成有关。

Abstract:Objective To investigate the 45% ethyl alcohol (EtOH) extracts from Xiaochaihu Tang on the proliferation of HepG2 human liver cancer cells and potential mechanisms. Methods ATP-Lite assay was ,performed to test the proliferation of HepG2 liver cancer cell line and antioxidant capacity was measured by oxygen radical absorbance capacity (ORAC). The effects of Xiaochaihu Tang on nitric oxide (NO) production were tested by LPS-aetivated RAW 264.7 routine rnacrophages using Griess reaction. Results The 45% EtOH Xiaochaihu Tang extracts displayed a cytotoxic response on HepG2 cells at 62.5± 500 mg/L, dose-dependently. The 50 percent effective concentration (ICso) was (90.7±23.7)mg/L. The ORAC value of the Xiaochaihu Tang was 1.3 times of Vitamin C. The 45% EtOH extract had significant inhibitory effect on NO production at 3.125±50 mg/L (P〈0.05, P〈0.01). Conclusion The extracts of Xiaochaihu Tang have in vitro antiproliferative effect on HepG2 cell, which may related with its antioxidant and anti-inflammatory activity.
