摘    要:目的评价注射用法罗培南的体内抗菌活性。方法建立小鼠腹膜炎模型,比较注射用法罗培南及厄他培南对产酶的大肠埃希菌、肺炎克雷伯菌及对甲氧西林敏感的金黄色葡萄球菌、耐万古霉素的粪肠球菌感染小鼠的体内抗菌活性。结果注射用法罗培南对产超广谱β内酰胺酶(ESBLs)的大肠埃希菌、肺炎克雷伯菌和对甲氧西林敏感的金黄色葡萄球菌有较强的体内和体外抗菌活性,其ED_(50)分别为10.96、9.12和7.07 mg·kg~(-1),厄他培南则为14.84、12.58和10.16 mg·kg~(-1)。法罗培南对耐万古霉素的粪肠球菌体外敏感,而厄他培南显示耐药。法罗培南对耐万古霉素的粪肠球菌的ED_(50)比厄他培南低66.77%,分别为10.75和32.35 mg·kg~(-1)。结论注射用法罗培南和厄他培南对产ESBLs的大肠埃希菌、肺炎克雷伯菌和对甲氧西林敏感的金黄色葡萄球菌有较强的体内和体外抗菌活性,两者作用相似。法罗培南对耐万古霉素的粪肠球菌感染可能有治疗作用,效果优于厄他培南。

Abstract:AIM To evaluate the antibacterial activities of faropenem for injection in vivo.METHODS Mice peritonitis models were established by using beta-lactamase producing strains Escherichia.coli,Klebsiella. pneumoniae and methicillin-sensitive Staphylococcus.aureus(MSSA),vancomycin-resistant Enterococcus. faecalis(VRE).The trial products including faropenem and ertapenem for injection were given intramuscularly and then compared their antibacterial activities.RESULTS The antibacterial activities of faropenem for injection in vivo and in vitro against beta-lactamase producing strains Escherichia.coli,Klebsiella.pneumoniae and MSSA were strong with ED_(50)as 10.96,9.12 and 7.07 mg'kg~(-1)respectively;comparatively for ertapenem,as 14.84,12.58 and 10.16 mg.kg~(-1)respectively.VRE was drug susceptible to faropenem but resistant to ertapenem in vitro.ED_(50)of faropenem was 66.77 % lower than ertapenem(10.75 and 32.35 mg·kg~(-1)).CONCLUSION The antibacterial activities of faropenem for injection in vivo and in vitro against beta-lactamase producing strains Escherichia.coli,Klebsiella.pneumoniae and MSSA are strong and similar to ertapenem.Faropenem for injection may possess the effect to treat infection of VRE,better than ertapenem.
