三七总皂苷伍用淫羊藿苷改善血管性痴呆大鼠空间学习记忆及抗脂质过氧化研究

目的:研究三七总皂苷(PNS)伍用淫羊藿苷(ICA)对血管性痴呆大鼠学习记忆、血液流变学及脑组织脂质过氧化的影响。方法:SD大鼠给药组各10只,阳性药组10只,采用反复夹闭双侧颈总动脉(CCA)再灌注同时腹腔注射硝普钠方法建立血管性痴呆大鼠模型,连续给药21天后,用跳台法判断给药前后大鼠的空间学习和记忆能力,再进行血液流变学和脑组织SOD、MDA的测定。结果 :与假手术组大鼠相比,模型大鼠跳台学习和记忆错误次数明显增加(P<0.05),大鼠造模前后连续口服三七总皂苷伍用淫羊藿苷2.5+20、5+40、10+80 mg/kg各剂量组21d后,大鼠上述行为学指标得到明显改善(P<0.05),全血粘度得到明显改善(P<0.05),脑组织SOD活性提高(P<0.05)和MDA含量降低(P<0.01)。结论:三七总皂苷伍用淫羊藿苷对血管性痴呆大鼠空间学习和记忆障碍有显著的预防和治疗作用,可能与通过改善血管性痴呆大鼠大脑血液供应和自由基代谢有关,从而起到保护大脑的作用。

Effects of icariin combined with Panax notoginseng saponins on vascular dementia model rats and anti lipid peroxidation in brain

Objective:To observe the effects of PNS and ICA on learning and memory deficit,blood viscosity and lipid peroxidation of brain in vascular dementia rats.Methods: 61 mice were randomly divided into 6 groups.Each of the rats was repeatedly made ischemia and reperfused for two times,combined with intraperitoneal injection of sodium nitroprusside,to establish vascular dementia models.Special learning and memory ability were evaluated by step-down passive avoidance test after treated 21 date,and biochemical estimations and blood viscosity measurement were carried.Results :The learning and memory abilities of model group rats significantly weakened with those in the saline group through step-down passive avoidance test(P<0.05).The combination significantly protected and improved all the evaluated indexes of the model groups(P<0.05),moreover,reduced the blood viscosity(P<0.05),increased the activity of SOD(P < 0.05),and decreased the contents of MDA(P<0.01).Conclusion: The combination can prevent and improve VD model rats,which is related with blood viscosity and the oxidative stress parameters i.e.levels of MDA,SOD in the brain.
