乌头碱对斑马鱼心脏毒性的初步研究

摘    要:目的:观察不同浓度的乌头碱对斑马鱼心脏的影响。方法:以发育正常的受精后48h(48hpf)的斑马鱼胚胎作为心脏毒性模型,以不同浓度的乌头碱处理上述胚胎,分别于处理后12h、24h观察胚胎心脏的形态和心率的变化。结果:5mg/L乌头碱作用24h未引起胚胎心脏中毒,而10、30、60mg/L乌头碱均导致胚胎心脏中毒,出现心膜出血,血细胞在心区堆积,心包囊水肿等现象。处理12h后心率随着浓度的升高而升高,且随后随着时间的延长心率下降。乌头碱引起斑马鱼胚胎心脏毒性的EC50(24h)约为14.49mg/L。结论:乌头碱对斑马鱼胚胎有心脏毒性作用,且与乌头碱的浓度正相关。

Abstract:Objective: To investigate the effect of Aconitine of various concentration on the heart of the zebrafish embryo.Methods: The zebrafish embryo 48h after fertilization with normal development was employed to test the cardiac toxicity of Aconitine of various concentration;By exposing the embryo to Aconitine of various concentration for 12h and 24h respectively,the change of morphology and heart rate were documented.Results: exposure to Aconitine at concentration of 5mg/L did not produce cardiac toxicity of the zebrafish embryo,whereas exposure to it at concentration of 10 mg/L、30 mg/L and 60mg/L all resulted in cardiac toxicity of the zebrafish embryo,leading to symptoms such as endocardium bleeding and aggregation of blood cells in the chambers as well as swelling of the heart.The heart rate is proportionally related to the concentration after exposure to Aconitine for 12h,after which the heart rate decreased.The EC50(24h) of the cardiac toxicity of Aconitine to zebrafish embryo is 14.49mg/L.Conclusion: Aconitine has cardiac toxicity to zebrafish embryo.The cardiac toxicity of Aconitine is dose and time-dependent.
