摘    要:目的:探讨高糖对人骨骼肌细胞株(HSkMCs)线粒体融合蛋白2(mfn2)表达及细胞凋亡的影响。方法:体外培养HSkMCs细胞株,将细胞分别以5.55 mmol/L,11.10 mmol/L,22.20 mmol/L葡糖糖培养48 h,检测mfn2mRNA表达及细胞凋亡情况。RT-PCR法测定基因表达,流式细胞技术检测细胞凋亡。结果:以5.55 mmol/L葡萄糖组培养48 h为基础对照,11.10 mmol/L组mfn2表达较对照组下降12.3%(P<0.05);22.20 mmol/L组mfn2表达较对照组降低47.3%(P<0.05);对照组细胞凋亡率为(1.80±0.57)%,11.10 mmol/L葡萄糖组细胞凋亡率为(13.23±1.47)%,22.20 mmol/L葡萄糖组细胞凋亡率为(24.03±3.05)%,均较对照组显著增加(P<0.05)。结论:高糖可诱导HSkMCs细胞mfn2 mRNA表达下调,增加其凋亡。

Effects of High Glucose on Expression of Mitofusin-2 and Apoptosis in Human Skeletal Muscle Cells

Abstract:Objective To investigate the effect of high glucose on the expression of mitofusin-2(mfn2) mRNA and apoptosis in human skeletal muscle cells.Methods HSkMCs were cultured and treated with different concentrations of glucose(5.55,11.10,22.20 mmol / L) for 48 h,respectively.Expression of mfn2 mRNA was detected by RT-PCR.Apoptosis was studied by flow cytometry.Results Compared with the expression of mfn2 mRNA in HSkMCs cultured with 5.55 mmol / L glucose for 48 h,the expression of mfn2 in 11.10 mmol / L group was significantly decreased by 12.3%(P < 0.05),in 22.20 mmol / L group deceased by 47.3%(P < 0.05).The apoptosis of HSkMCs cultured with 5.55,11.10,22.20 mmol / L glucose groups was(1.80 ±0.57) %,(13.23 ±1.47) %,(24.03 ± 3.05) %,respectively,and there was a significant difference(P < 0.05).Conclusion High glucose can downregulate the expression of mfn2 mRNA and induce the apoptosis of HSkMCs.
