羟基红花黄色素A抑制新生血管形成的机制研究

摘    要:目的从中药中寻找新的血管生成抑制剂,并探讨其抑制新生血管生成的作用机理.方法从红花中提取分离羟基红花黄色素A,利用鸡胚尿囊膜(CAM)实验观察羟基红花黄色素A对新生血管的抑制作用,并通过RT-PCR实验,观察羟基红花黄色素A对CAM组织中碱性成纤维细胞生长因子(bFGF)、血管内皮细胞生长因子(VEGF)及其受体(VEGF-R)(flt-1) mRNA表达的影响.结果提取出的羟基红花黄色素A纯度达到了98.9%,浓度为0.59 g/L的羟基红花黄色素A能使CAM血管数明显变细减少(P<0.01)并能显著抑制CAM组织中bFGF、VEGF及VEGF-R(flt-1) 的mRNA表达(P<0.05).结论通过本实验我们首次得出羟基红花黄色素A能显著抑制CAM新生血管生成,其作用机制之一是通过抑制bFGF、VEGF及VEGF-R(flt-1)的mRNA表达来实现的,提示羟基红花黄色素A可能是一很有潜力的血管生成抑制剂.

Abstract:Objective To find new angiogenic inhibitors from TCM drugs and investigate their mechanisms of inhibiting angiogenesis.Methods Hydroxysafflor yellow A was separated from the dried flowers of Carthamus tinctorius; the chick embryo chorioallantoic membrane (CAM) test was carried out to observe the effect of hydroxysafflor yellow A on angiogenesis; RT-PCR was used to observe the effects of hydroxysafflor yellow A on the mRNA expressions of bFGF, VEGF and VEGF-F(flt-1) in the CAM.Results Hydroxysafflor yellow A extracted from the dried flowers of Carthamus tinctorius was of purity of 98.%; hydroxysafflor yellow A could markedly reduce the number of CAM vessels when it was at the concentration of 0.59 g/L (P<0.01), and remarkably inhibit the mRNA expressions of bFGF, VEGF and VEGF-F(flt-1) in the CAM (P<0.05). Conclusion It is the first time when hydroxysafflor yellow A has been discovered to have the effects of markedly inhibiting angiogenesis in CAM, one of the mechanisms of which may be its inhibiting the mRNA expressions of bFGF, VEGF and VEGF-F(flt-1). The results of the experiment suggest that hydroxysafflor yellow A may become a potential angiogenic inhibitor.
