舒芬太尼对体外循环大鼠脑损伤的保护作用

摘    要:
目的:观察舒芬太尼对体外循环(CPB)大鼠的脑保护作用。方法:成年雄性SD大鼠24只,随机分为4组,每组6只:假CPB组(Sham组)、CPB组、1μg/Kg舒芬太尼组(S1组)和5μg/Kg舒芬太尼组(S5组)。所有动物麻醉后机械通气。Sham组进行相关手术和插管但不行CPB;余下各组均参照有关方法改良后建立CPB。S1组和S5组于CPB开始前转流管道中分别给予负荷剂量的舒芬太尼1μg/Kg和5μg/Kg,随后分别用维持剂量90μg/Kg·min和300μg/Kg·min直到CPB后1h。4组大鼠颈静脉取血后断头处死,取脑组织,测其含水量和总钙水平,以及血清S100β含量。结果:与Sham组比较,CPB组脑组织含水量、总钙水平及血清S100β含量均明显升高(P0.05);与CPB组比较,S1组和S5组脑组织含水量、总钙水平及血清S100β含量均明显降低(P0.05),且S1组下降多于S5组,尤其脑组织含水量明显低于S5组(P0.05)。结论:应用舒芬太尼可以减轻CPB大鼠脑损伤,以1μg/Kg剂量为宜。

Protective Effects of Sufentanil on Cardiopulmonary Bypass Rats' Brain Injury
Abstract:
Objective:To observe protective effects of Sufentanil pretreatment on cardiopulmonary bypass(CPB)rats' brain.Method:24adult male SD rats were divided into 4groups randomly,i.e.,Sham CPB group(Sham group),CBP group,1μg/Kg Sufentanil pre-treatment group(S1group)and 5μg/Kg Sufentanil pre-treatment group(S5group),and each group was made up of 6rats.After all animals had been narcotized,mechanical ventilation was carried out.For the Sham group,relative operations and intubation tubes were implemented,but CPB was not carried out.As for the other groups,CPB was established after improvement had been performed according to Wang M method.In S1and S5groups,1μg/Kg and5μg/Kg Sufentanil at a load dosage had been given in a shunt before CPB was started,respectively.Later,90μg/Kg min and 300μg/Kg min doses were maintained until 1hafter CPB,respectively.After blood was got from four groups of rats' jugular vein,brain tissue was taken.In this way,its total calcium level,water content and content of serum S100βwere measured.Results:Compared with the Sham group,the water content,the total calcium level and the content of serum S100βof brain tissue in the CPB group were obviously raised(P<0.05);compared to the CPB group,the total calcium level and the content of serum S100βof brain tissue in the S1and S5groups were significantly reduced(P<0.05),and the reduction of the S1group was higher than that in the S5group,especially the water content of brain tissue in the S1groups was obviously lower than that in the S5group(P<0.05).Conclusion:Sufentanil pretreatment could reduce CPB rats' brain injury and 1μg/Kg dose was the most appropriate.
