谷氨酸转运体在体外培养大鼠皮质星形胶质细胞机械性损伤中的表达变化

摘    要:【目的】研究机械性损伤后星形胶质细胞内谷氨酸转运体的表达变化,并探讨其在细胞损伤过程中可能的作用机理。【方法】采用免疫组化方法对培养的细胞进行鉴定,流式细胞术和Western Blotting定量谷氨酸转运体蛋白的表达改变。【结果】流式细胞术结果表明,机械性划伤12h和24h后,谷氨酸转运体(EAATl)蛋白表达均较对照组显著增加(P〈0.05),而损伤12h和24h组组间无显著差异(P〉0.05);Western blotting显示,对照组、机械性划伤12h和24h组谷氨酸转运体(EAATl)蛋白表达吸光度(A)相对值为:0.21、0.44和0.46。与对照组比较,损伤后12h和24h星形胶质细胞EAATl的蛋白表达均明显升高(P〈0.05),损伤两组间表达无显著性差异(P〉0.05)。【结论】机械性损伤可导致星形胶质细胞内EAATl的表达明显升高,提示其可能在脑损伤的发生发展以及修复过程中起到了一定的作用。

Abstract:[Objective] To investigate the changes of glutamate transporters expression after mechanical injury, and analyze the possible mechanism. [Methods] To identify and quantitate astrocyte and glutamate transporters changes in vitro using immunohistochemical staining flow cytometry and western blotting. [Results] Flow cytometry showed expression of EAAT1 increased postinjury 12 h and 24 h, as compared with control (P 〈 0.05). Western blotting showed the absorbance (A) of 12 h and 24 h after injury was increased as compared with control group (P 〈 0.05). Two injury groups did not show obvious differentce in expression. [Conclusions] Mechanical injury could result EAAT1 expression increase in astrocyte, and EAAT1 may have have some effect during brain injury.
