曲尼司特对TGF-β1诱导的NRK-52E细胞E—cadherin表达的影响

摘    要:
目的观察转化生长因子β1(TGF—β1)诱导的NRK-52E细胞转分化过程中转分化标志物E-钙黏蛋白(E—cadherin)的表达情况及曲尼司特对其转分化过程的影响。方法应用TGF—β1(8mg/L)刺激NRK-52E细胞,分别分为阴性对照组、TGF—β1诱导组和不同剂量曲尼司特干预组,采用流式细胞术和蛋白质印迹检测各组E—cadherin的表达情况。结果流式细胞术和蛋白质印迹检测结果表明,TGF—β1刺激后,NRK-52E细胞E—cadherin的表达均显著减低。结论曲尼司特能够抑制TGF—β1诱导的NRK-52E转分化,其机制可能与上调E—cadherin的表达有关。

Abstract:
Objective It is to observe the expression of transdifferentiation marker E - cadherin during transdifferentiation of NRK - 52E cells induced by TGF - 1, and the effect of tranilast on this transdifferentiation. Methods The NRK - 52E cells cultured with TGF- 18 mg/L were divided into five groups: the negative control group, TGF- 1 induced group, and different dosage of tranilast intervention groups. The expressions of E - cadherin protein in every group were detected by flow cytometry and western blot. Results Flow cytometry and western blot analysis showed that the expression of E - cadherin was dramatically decreased in response to TGF - 1 treatment in NRK -52E cells. Conclusion Transdifferentiation of cultured NRK - 52E induced by TGF - 1 can be inhibited by certain level of tranilast, which mechanism may be related to its promoting effect on the expression of E - cadherin.
