黄芪对单侧输尿管梗阻大鼠肾小管间质病理损害的作用

摘    要:目的观察黄芪(AM)对单侧输尿管梗阻(UUO)所致大鼠肾小管间质病理损害的影响,初步探讨黄芪拮抗纤维化的作用机制。方法48只SD大鼠随机分为假手术(Sham)、UUO组和UUO+黄芪治疗组(AM)、UUO+氯沙坦治疗组(ARB)。在造模后7、14天处死动物,观察HE染色、Masson染色和PAS染色肾脏病理改变,并对肾小管间质损害进行半定量评定分析。结果与假手术组相比,UUO术后大鼠肾脏病理损害进行性加重,肾小管损伤程度、肾间质相对面积和肾小管萎缩程度评分均明显增高,黄芪干预后各时间点上述指标明显改善,与ARB作用相似。结论黄芪能有效抑制肾小管间质病理损害,减轻和改善UUO大鼠肾间质纤维化。

Effect of Astragalus mongholicus on pathological lesion of renal tubulointerstitium in SD rats with unilateral ureteral obstruction

Abstract:Objective To study the effect of Astragalus mongholicus (AM) on renal fibrosis in SD rats with unilateral ureteral obstruction (UUO). Methods Forty-eight Sprague-Dawley rats were randomly divided into 4 groups: sham-operation group(Sham),UUO group (UUO), UUO + AM group (AM) and UUO + losartan group (ARB). Animals in ARB group were intragastrically administrated with losartan 20 mg·kg^-1·d^-1 after UUO operation, while those in Sham, UUO and AM groups were intragastrically administrated with identical voluminal normal saline. AM group was intraperitoneally injected with astragalus mongholicus 10 g·kg^-1·d^-1, while Sham, UUO and ARB groups were intraperitoneally injected with identical voluminal normal saline. At 7, 14 days after UUO, the dynamic histological changes of renal interstitial tissues were observed and renal damage including tubular lesions, interstitial fibrosis and tubular atrophy were quantified on HE, Masson and PAS stained tissue sections. Results Renal damage was exacerbated and the sores of pathological lesions were all significantly increased in UUO group compared with those of Sham group(P 〈0. 05)at each time point. Tubular lesions, interstitial fibrosis and tubular atrophy were significantly alleviated by AM treatment (P〈0. 05),and there was no difference between AM and ARB groups (P〉0. 05). Conclusion AM can improve renal tubulointerstitial damage and ameliorate renal interstitial fibrosis induced by UUO in rats.
