乳腺癌转移相关基因表达蛋白组织微阵列的初步研究

摘    要:目的:通过MMP2、P27Kipl、纤维连接蛋白(fibronectin)、MDM2、P21WAF1免疫组化染色,分析上述不同蛋白在乳腺癌组织中的表达,探讨乳腺癌转移相关分子生物学机制.方法:收集具有淋巴结转移的乳腺癌病例100例,制作成组织芯片,进行MMP2、P27Kipl、fibronectin、MDM2、P21WAF1免疫组化染色,根据染色指数(staining index,阳性细胞百分数×染色强度)对乳腺癌原发灶和转移灶进行统计学处理,分析上述不同指标在乳腺癌转移中的作用.结果:100例乳腺癌标本中,转移灶MMP2、P27Kipl、fibronectin、MDM2、P21WAF1的表达均高于原发灶肿瘤组织的表达,其中转移灶MMP2、P27Kipl、fibronectin、P21WAF1的表达与原发灶的差别具有统计学意义,P<0.05.结论:MMP2、P27Kipl、fibronectin、MDM2、P21WAF1表达相关基因可能参与乳腺癌转移,肿瘤细胞表达MMP2、P27Kipl、fibronectin、MDM2、P21WAF1蛋白增加,可能更易于其转移和浸润.从另外一个侧面说明:在乳腺癌演进的过程中,肿瘤细胞获得侵袭能力可能不是由单个基因进行调控的,而是多个基因相互协调作用的结果.

Abstract:Objective: To analyze different antigen expression in breast cancer and investigate correlative mole-biomechanism the immunohistochemical detection of MMP2?P27Kipl?fibronectin?MDM2?P21WAF1 were performed. Methods: To collect 100 cases of breast cancer with lymph node or soft tissue metastasis and crank out tissue microarray with these tumor. we detected these tumor tissue with immunohistochemistry stain on tissue microarray sections for MMP2?P27Kipl?fibronectin?MDM2?P21WAF1. According to staining index that is product of positive cells percent and positive degree the statistical analysis was put up between the primary and the metastasis of breast cancer to assay different marker in the procession of breast cancer metastasis. Results: In the 100 cases breast cancer MMP2?P27Kipl?fibronectin?MDM2?P21WAF1 expression metastasis focus is higher than primary focus. The different is significance in stastasis other than MDM2. Conclusion: The genes about MMP2?P27Kipl?fibronectin?MDM2?P21WAF1 maybe take part in the procession of breast cancer pervasion. The high expression of MMP2?P27Kipl?fibronectin?MDM2?P21WAF1 maybe make tumou cells for metastasis and invasion. Also it shows that multi-genes, instead of monogenes, participate the regulation of the malignant actions.
