肾康丸对早期糖尿病肾病大鼠足细胞损伤的保护作用

摘    要:目的 观察肾康丸对糖尿病肾病(DN)大鼠肾小球足细胞形态及desmin、podocin表达的影响,探讨肾康丸对DN的肾保护作用机制.方法 应用链脲佐菌素建立大鼠DN模型,将DN模型大鼠随机分为4组:DN模型对照组、肾康丸组、厄贝沙坦组、肾康丸和厄贝沙坦合用组;另设正常对照组.各组分别干预8周后,观察24h尿蛋白定量变化,利用光镜、电镜观察肾脏病理改变,应用免疫组化技术观察足细胞desmin、podocin表达变化.结果 应用肾康九、厄贝沙坦干预后,DN大鼠24 h尿蛋白定量明显减少,肾脏病理改变显著减轻,足细胞desmin表达明显减少、podocin表达明显增加.结论 通过上调足细胞podocin的表达,使podocin的合成增加,从而减轻足细胞的损伤、维护足细胞结构和功能的完整性,可能是肾康丸对DN的肾保护作用机制之一.
