| 摘 要： | 目的:探讨H2O2是否通过调节P38MAPK的活性而诱导PC12细胞凋亡。方法:碘化丙啶(PI)染色流式细胞术(FCM)检测细胞凋亡;Western-Blot测定磷酸化ERK1/2蛋白和磷酸化p38蛋白的表达。结果:作用PC12细胞24h后,20～80μmol/L的H2O2可呈浓度依赖性地诱导PC12细胞凋亡并增加PC12细胞P38磷酸化的水平;P38特异性抑制剂SB203580(10或20μmol/L)预处理30min可显著减轻40μmol/LH2O2对PC12细胞凋亡的诱导作用。结论:H2O2通过上调P38的活性而诱导PC12细胞凋亡。 |

| Abstract: | Objective To investigate whether hydrogen peroxide(H2O2) -induced apoptosis of PC12 cells is involved in regulation of P38-MAPK.Methods The apoptosis of PC12 cells was detected by flow cytometry(FCM) with propidium iodide staining;the expression levels of P38 were analyzed by Western blotting.Results Treatment of PC12 cells with H2O2 at the concentration of 20,40,80μmol/L for 24 h induced apoptosis of PC12 cells and increase in activation of P38 in a concentration-dependent manner.Pretreatment with SB203580(10 and 20 μmol/L) for 30 min decreased the apoptosis of PC12 cells induced by 40μmol/L H2O2 for 24 h.Conclusion Hydrogen peroxide induce apoptosis of PC12 cells via up-regulating P38-MAPK pathway. |
