摘    要:
目的:探讨去铁胺(DFO)对急性心肌缺血大鼠心肌梗死面积、心肌血管新生及心肌组织血管内皮生长因子(VEGF)表达的影响.方法:健康SD大鼠随机分为4组:正常对照组(Nor组)、假手术组(Sham组)、心肌梗死+生理盐水组(MI组)、心肌梗死+DFO组(DFO组).开胸手术结扎冠状动脉前降支制作心肌梗死模型(Sham组只穿线不结扎),DFO组和MI组术前24 h分别给予DFO (200 mg/kg)或等体积生理盐水腹腔注射.TTC染色后检测心梗面积;RT-PCR检测梗死边缘区心肌VEGF的表达;免疫组化法检测梗死边缘区心肌微血管密度及VEGF蛋白的表达.结果:DFO组心梗面积明显低于MI组,梗死边缘区微血管密度高于MI组、Sham组和Nor组.MI组心肌组织VEGF蛋白表达明显比Nor组和Sham组高;DFO组心肌组织VEGF蛋白比MI组表达进一步上调.VEGF mRNA的表达与VEGF蛋白表达变化一致.结论:DFO可以减少急性心肌缺血大鼠心梗面积,促进缺血区域血管新生,其作用机制与上调VEGF的表达有关.

Abstract:
Objective To investigate the effects of desferrioxamine (DFO) on myocardial infarct size, myocardial neovascularization and the expression of vascular endothelial growth factor (VEGF) in acute myocardial infarction (AMI) rats. Methods Healthy Sprague-Dawley rats were randomly divided into 4 groups: Nor group, sham group, MI group and DFO group. Seven days after surgery, the myocardial infarct size was detected by TTC staining. The expression levels of VEGF mRNA and protein and microvessel density (MVD) in the marginal zone of myocardial infarction were determined by RT-PCR and immunohistochemical method, respectively. Results The myocardial infarct size in DFO group was significantly lower than to that in MI group. The MVD of the marginal zone of myocardial infarction in DFO group was significantly higher than that in MI, sham and Nor groups. Compared with Nor group and sham group, the expression levels of VEGF mRNA and protein were significantly increased. Compared with MI group, the VEGF expression and MVD in DFO group were significantly increased. Conclusion DFO may reduce the myocardial infarct size and induce angiogenesis in ischemic regions via up-regulating the VEGF expression in AMI rats.
