葛根素对糖尿病大鼠主动脉硫酸乙酰肝素蛋白多糖影响的实验研究

摘    要:目的 观察葛根素对糖尿病大鼠主动脉HSPG的影响。方法 利用链脲佐菌素腹腔注射法诱导建立1型糖尿病大鼠模型,将实验用SD大鼠随机分正常对照组、糖尿病组、葛根素治疗组。治疗16周,观察治疗期间及治疗后大鼠的一般状况、血糖(BS)、三酰甘油(TG)、胆固醇(TC)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、糖化血红蛋白(HbA1c)、糖化低密度脂蛋白(G-LDL),分离主动脉,HE染色及免疫组织化学检测主动脉内膜HSPG表达。结果 造模2组大鼠均出现血脂异常及主动脉病理形态改变。葛根素能改善基本状况,葛根素能降低糖尿病大鼠的三酰甘油(P〈0.05)、胆固醇(P〈0.05)、低密度脂蛋白(P〈0.05)、糖化血红蛋白(P〈0.05)、糖化低密度脂蛋白(P〈0.05),升高高密度脂蛋白(P〈0.05);葛根素增强主动脉硫酸肝素蛋白(P〈0.05)作用。结论 葛根素可以通过增强硫酸肝素蛋白对糖尿病主动脉具有保护作用。

Abstract:[Objective] To observe the effect of Puerarin aorta expression of HSPG in Streptozotocin-induced diabetic rats [Methods] Diabetic nephropathy rats were induced by intraperitoneal injection of STZ. Rats were allocated to normal control group, model group, and Puerarin group for 16 weeks. During and after the treatment, its general state, blood suger (BS), triglyceride (TC), cholesterol, low density lipoprotein (LDL-C), high density lipoprotein (HDL-C), glycosylated hemoglobin (HbA1c) and glycosylated low density lipoprotein (G-LDL) alteration of tissue morphology via H. E staining was observed after the aorta was taken out from the rats executed by narcosis. Heparin sulfate proteoglycan protein, were determined by immunohistochemistry method. [Results] Diabetes mellitus and aorta lesion occurred in the tow model groups. Puerarin could improve the general state, improve the general state, decrease triglyceride (P <0.05), serum cholesterol (P <0.05), low density lipoprotein (P <0.05), glycosylated hemoglobin (P <0.05), and glycated low density lipoprotein (P <0.05), increase high density lipoprotein (P <0.05). The expression of HSPG in the kidney was significantly inhibited (P <0.05) at the same time [Conclusion] Puerarin has a certain effect in preventing aorta by incteasing expression of heparan sulfate proteoglycan.
