葛根素对代谢综合征-2型糖尿病大鼠胰岛素、血糖、血脂的影响掌

摘    要:目的观察葛根素对代谢综合征-2型糖尿病大鼠(MS—T2DM)的胰岛素敏感性及血糖、血脂和氧化应激的影响。方法采用腹腔注射小剂量链脲霉素25~30mg/(kg·d),每日1次,并加饲高热量饮食(富含脂肪和蔗糖)42d,造成糖耐量减退、胰岛素抵抗(IR)的MS—T2DM大鼠模型;葛根素0.3g/kg、0.15g/kg或二甲双胍0.25g/kg灌胃给药,每日1次,给药28d;测病鼠血糖、血脂、胰岛素(Fins)和丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性,计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(IRI)。结果葛根素和二甲双胍均能显著降低病鼠Fins和IRI,而提高ISI(P〈0.01),增强胰岛素敏感性;降低FPG和口服葡萄糖耐量试验后2h血糖(OGTr-2hPG)水平,以及降低总胆固醇(TC)、低密度脂蛋白胆固醇(LDL—C)、高密度脂蛋白胆固醇(HDL—C)、甘油三酯(TG)、游离脂肪酸(FFA)、糖化血红蛋白(GHb)、果糖胺(FRA)含量(P〈0.01),改善糖脂代谢紊乱;降低MDA含量和提高SOD活性(P〈0.01),增强清除自由基能力。结论葛根素对MS—T2DM大鼠能产生二甲双胍样的改善高胰岛素血症、提高胰岛素敏感性以及降糖和调节血脂等效应,并具有抗氧化活性。

Abstract:Objective To investigate the effects of puerarin on insulin sensibility and oxidative stress-free radical injure in metabolic syndrome-type 2 diabetic rats (MS-T2DM). Methods The model with lower dosage of streptozocin (25- 30 mg/(kg·d), intraperltoneally injected, q, d.,1 d) and high calorie diet (abounded with sugar and fat) fed for 42d,and the model of MS-T2DM with insulin resistance were performed; the effects of puerarin (0.3 g/kg, 0. 15 g/kg intragastrointestinally treated, q. d,,28 d) on the levels of fasting blood glucose (FPG), 2 h blood glucose (2h PG) after oral glucose tolerance test (OGTF) in vivo, and serum levels of total cholesterols (TC), low density lipoprotein - cholesterol (LDL - C), high density lipoprotein - cholesterol (HDL - C), triacylglyeerol (TG), free fatty acid (FFA) fruetosamine (FRA), Glyeosylated hemoglobin (GHb), fasting insulin (Fins), malondialdehyde (MDA), insulin resistant index (IRI) and insulin sensitivity index (ISI), the activity of superoxide dismutase (SOD) in MS-T2DM with impaired glucose tolerance (IGT) were studied. Results Puerarin reduced the levels of FPG and 2 h PG after OGTT (P 〈 0.01) and improved IGT in MS-T2DM, and the contents of MDA, TC, LDL - C, HDL - C, TG, FFA, Fins were lowered ( P 〈 0.01 ) and IRI was very significantly decreased ( P 〈 0.01 ), ISI was enhanced (P 〈 0. 01) and the activity of SOD was heightened (P 〈 0.01), compared with the models of MS-T2DM. Conehlsion Puerarin can lower hyperinsulinemia, ameliorate insulin sensibility, reduce blood glucose and lipids, and which mechanism is similar to the kind of metformin, and potentiates the effect of antioxidation in MS- T2DM.
