| 摘 要： | 以不同技术路线制备Ｐ２１０／ＭＭＣ，Ｐ２１０／ＴＮＰ及Ｐ４０／ＣＤ３三种双功能抗体（ＢｓＡｂ）。在１１次细胞融合中，接种７４８４孔，获１５株二次杂交瘤，其中５株分泌ＢｓＡｂ。将亲本杂交瘤分别诱变为ＨＧＰＲＴ＾－及ＴＫ＾－，融合后以ＨＡＴ选择培养并辅以饲养细胞较易获得二次杂交瘤。证明两种抗原交互反应的桥联试验，特别是当二次杂交瘤分泌不同类别的Ｉｇ时，对于确证ＢｓＡｂ的存在有着重要意义。本文首次报告，

| Abstract: | Bispecific monoclonal antibodies (BsAb) P210/MMC P210/TNP and P40/CD3 were prepared with different technical approaches. Among 11 fusions, 7484 wells were planted, and 15 hybrid-hybridomas were obtained, from which only 5 secreted BsAb. It was more successful to get hybrid-hybridomas,when the parental hybridomas were induced to HGPRT- and TK- mutants for fusion and feed cells were used in HAT selective cultures. In addition,it was demonstrated that bridge assay was significant to identify the characters of BsAb,especially when hybrid-hybridomas secreted different classes of Ig. In present study,it was reported that the frequency of BsAb recombined from r2b and u was about 16. 7% (2/12). |
