外周血中PCA3基因表达及其在前列腺癌诊断中的意义

摘    要:目的探讨PCA3对活检阴性的前列腺癌(Pca)临床诊断的应用价值研究。方法收集2011年01月至2012年12月间在我院就诊病理诊断为前列腺癌及前列腺增生患者。同时纳入健康的男性志愿者为正常对照。对所有监测对象,采用荧光定量PCR检测外周血PCA3 mRNA的表达。结果共纳入66例做为对象,其中前列腺癌患者26例,良性前列腺增生共30例,正常对照组10例。健康男性及前列腺增生患者外周血均无PCA3基因的表达,前列腺癌患者有21例表达,PCA3表达阳性率为80.8%(21/26)。前列腺癌组与健康组及增生组均有明显统计学差异(P〈0.01)。PCA3阳性率与PCa临床病理差异无统计学意义,但PCA3基因的表达与PCa临床病理参数差异有统计学意义。结论PCA3基因在外周血的mRNA表达在前列腺癌诊断上有良好特异性。

The expression of PCA3 gene in peripheral blood and the significance of PCA3 score for diagnosis of prostate cancer

Abstract:Objective To evaluate the application value of PCA3 in pathological study of the prostate cancer which the biopsy was negative. Methods 2011-01 to 2012-12 the patient in our hospital were diagnosed as prostate cancer and benign prostatic hyperplasia. While incorporating healthy male volunteers as normal controls. For all monitored objects, using quantitative PCR test the expression of PCA3 mRNA in peripheral blood. Results 66 cases were selected, of which a total of 26 cases of patients with prostate cancer, BPH total of 30 cases, 10 normal controls. Healthy males and patients with benign prostatic hyperplasia no PCA3 gene expression, there are 21 cases of prostate cancer patients expressing, PCA3 expression was 80.8% (21/26). Prostate cancer group and the healthy group and the group were statistically significant differences in proliferation (P〈0.01). PCA3 positive rate of PCa was no significant difference in clinical pathology, but the PCA3 gene expression and clinicopathological parameters PCa dif- ference was statistically significant. Conclusion PCA3 gene mRNA expression in peripheral blood in the diagnosis of prostate cancer have good specificity.
