高效液相色谱法测定氯霉素泡腾片中氯霉素含量

目的建立氯霉素泡腾片中氯霉素含量测定的高效液相色谱方法。方法采用高效液相色谱法,其中色谱柱为Kromasil 100-5 C18（250 mm×4.6 mm,5μm）;流动相为0.01mol/L庚烷磺酸钠缓冲溶液（取磷酸二氢钾6.8 g,用0.01 mol/L庚烷磺酸钠溶液溶解并稀释至1000 mL,再加三乙胺5 mL,混匀,用磷酸调pH值至2.5）-乙腈为70∶30;柱温为室温;检测波长为274 nm。结果氯霉素在20.7～72.6μg/mL范围内线性关系良好,平均回收率为100.10%,RSD值为0.93%。结论本法操作简便,专属性好,精密度高,可作为氯霉素泡腾片的含量测定方法。

Determination of Chloramphenicol in effervescent Chloramphenicol Tablets by HPLC

Objective To establish an HPLC method for the determination of Chloramphenicol in effervescent Chloramphenicol Tablets. Methods The column was Kromasil 100-5 C18（250 mm×4.6 mm, 5 μm） and 0.01 mol/L sodium heptane buffer（potassium dihydrogen phosphate 6.8 g, with 0.01 mol/L of sodium heptane was dissolved and diluted to1000 mL, plus triethylamine 5 mL, mixing, pH value adjusted to 2.5 with phosphoric acid）- acetonitrile（70∶30） was used as the mobile phase. The column temperature was room temperature, detection wavelength was 274 nm. Results The calibration curve was linear in the range of 20.7-72.6 μg/mL for Chloramphenicol. The average recovery was100.10% and RSD was 0.93%. Conclusion The method is simple, accurate and reliable, and can be used for the quality of effervescent Chloramphenicol Tablets.
