风疹IgG抗体ELISA检测方法的建立和应用

Establishment and Applicationtion of ELISA for the Detection of Anti - Rubella IgG

Objective:To establish an indirect Enzyme linked immunosorbent assay(ELISA) method for detecting rubella IgG antibody in serum and to evaluate its preliminary significance of application. Methods; The rubella antigen was extracted after succession of culture Vero cell infected with RV - GOS strain. An indirect ELISA was developed. Rubella IgG antibodies in serum specimen were detected in 270 women from 20 to 40 years old in baoan district of Shenzhen by the method established by our study and the ELISA assay kit provided by Diasorin Co. . Results: Compared 270 sample with ELISA assay of Diasorin Co. , the relative coefficient (r) was 0.92, P < 0.05, indicating a good relativity. There were 221 positive cases among 226 cases, resulting in a sensitivity of 97.79%. there were 42 negative cases among 44 cases, indicating a specificity of 95.45% , and concordance was 263 of 270 ( 97.41 % ). The intraassay coefficient of variation was between 7. 90 ~ 10. 67 % , and the inter-assay coefficient of variation was between 10. 63 ~ 16. 98%. Conclusion: Our ELISA assay is specific, sensitive, reliable, simple, and can be used screening the rubella antibody among premarital women.
