摘    要:目的 制备地榆鞣质提取物,观察其对白细胞减少症的影响.方法 采用明胶沉淀丙酮解析法制备地榆鞣质提取物.选用KM小鼠,按体重和性别随机分成空白组、模型组、阳性组(rhG-CSF组)和地榆鞣质高、中、低剂量(20、10、5 mg/kg)组,用环磷酰胺复制小鼠白细胞减少症模型,并以白细胞数(WBC)、脾脏系数和骨髓DNA含量作为检测指标,研究地榆鞣质的升白作用.结果 所制地榆鞣质纯度达90%;药理研究显示,地榆鞣质高、中、低剂量组均能显著升高白细胞数量(P< 0.05),且地榆鞣质高、中剂量组升高骨髓DNA的作用优于阳性药(rhG-CSF)组.结论 采用明胶沉淀丙酮解析制备法可获得纯度较高的鞣质部位,鞣质具有显著的升高白细胞及保护骨髓DNA的作用.

Abstract:Objective To prepare Burnet tannins extracts and observe its influence on Leukopenia. Methods Gelatin precipitation and acetone resolution methods were used to prepare Burnet tannins extracts. Mice of KM species were randomly classified into blank group, model group, positive group (rhG-CSF), and high, middle and low dosage (20, 10, 5 mg/kg) of tannins group, according to weight and gender. The leucopenia mice model was made by cyclophosphamide treatment, and white blood cells (WBC) number, spleen index and bone marrow DNA content were measured to test tannins' effect on the WBC. Results The purity of Burnet tannins extracts by this method reached about 90%. Pharmacological studies showed that WBC numbers in all of the three tannins groups can significantly be elevated (P 〈 0.05), and bone marrow DNA contents in tannins group of high and middle dosage (20 mg/kg, 10 mg/kg) are better than that in rhG-CSF group. Conclusions Higher purity of Burnet tannins can be extracted by method of gelatin precipitation and acetone resolution, and the Burnet tannins can significantly elevate WBC and protect bone marrow DNA.
