Abstract:Objective: To study the antibacterial activities of 9 traditional Chinese medicine monomers against Clostridium difficile in vitro. Methods: The antibacterial activities of 9 traditional Chinese medicine monomers against Clostridium difficile were screened by a filter paper method. For those with stronger activities, MICs were determined by an agar dilution method, MBCs were determined by a microbroth dilution method, and their antibacterial activity at pH 3-8 were determined by the filter paper method. A checker board method was used to determine their combined bacteriostatic effects with vancomycin. Results: Baicalein and quercetin had stronger antibacterial activities against Clostridium difficile, with MIC at 1.25 and 2.5 mg·mL-1, and MBC at 1.25 and 5 mg·mL-1, respectively. The antibacterial activities of baicalein and quercetin were stable in the pH range from 3 to 8. These two monomers showed synergistic antibacterial effects with vancomycin. Conclusion: Baicalein and quercetin had good antibacterial activities against Clostridium difficile in vitro, which provides a new idea for the treatment of Clostridium difficile infection.
